Air‐drying of human leucocytes for scanning electron microscopy using the GTGO procedure

Abstract: The utilization of tannic acid and guanidine hydrochloride as mordants for better osmium binding has been shown to serve as an excellent alternative to metal coating of organ tissue specimens for scanning electron microscopy (SEM). The present report describes the G T G O procedure, a modification of the TAO technique introduced by Murakami er al. (1977, 1978), which we have found successful for the Preparation of air dried peripheral blood leucocytes for SEM studies. Air dried, GTGO-treated leucocytes show ex… Show more

“…For SEM, 5 mL cultures were harvested and fixed with glutaric dialdehyde for 24 h. Then the cells were embedded using the glutaraldehyde, tannic acid, guanidine hydrochloride, osmium tetroxide (GTGO) technique [12], coated with gold, and examined under the SEM microscope (Quanta200; FEI, Hillsboro, OR, USA). For the TEM analysis, the harvested cells were fixed with 1% glutaric dialdehyde for 24 h, separated by centrifugation for 5 min at 1000× g, then washed five times with 0.1 M phosphate buffer.…”

“…These appearances, on the other hand, might be better ascribed to the mordant activity of TA, which has been related to non-polar van der Waals forces (Simionescu & Simionescu, 1976a, b;Bird, 1977;Singley & Solursh, 1980;Gamliel et al, 1983).…”

“…1968, Art. 773) and was used for EM preparations according to Gamliel et al (1983). However, when, hydrolysates of collagen-GAGs, which were obtained using crude TA, were subjected to amino sugar analysis (see below, protocol II, for the preparation of collagen-GAG complexes), the peaks of both N-acetylglucosamine (GlcNAc) and GalNAc were found to have been overridden by contaminating peaks.…”
