AID-Driven Deletion Causes Immunoglobulin Heavy Chain Locus Suicide Recombination in B Cells

Abstract: Remodeling of immunoglobulin genes by activation-induced deaminase (AID) is required for affinity maturation and class-switch recombination in mature B lymphocytes. In the immunoglobulin heavy chain locus, these processes are predominantly controlled by the 3' cis-regulatory region. We now show that this region is transcribed and undergoes AID-mediated mutation and recombination around phylogenetically conserved switchlike DNA repeats. Such recombination, which we term locus suicide recombination, deletes the … Show more

“…In parallel, CSR drives apoptosis of undesirable B cell clones through deletion of the Ig heavy chain in a process called locus suicide recombination. This prevents BCR expression and thus eliminates the survival signals the BCR transmits, inducing apoptosis [35]. Having survived selection in the light zone, GC B cells can do one of three things: they can re-enter the dark zone for additional rounds of proliferation and SHM [36,37].…”

“…Although their levels are low, the 3′RR transcripts [3′RR enhancer RNAs (eRNAs)] can readily be detected upon stimulation of splenic B cells and correlate with the 3′RR activity in activated B cells (23,24). Additionally, recent work involved hs4 eRNA in long-range interactions with a far downstream nonIg sequence (25).…”
