AGR2 Is a Novel Surface Antigen That Promotes the Dissemination of Pancreatic Cancer Cells through Regulation of Cathepsins B and D

Abstract: Pancreatic ductal adenocarcinoma (PDAC) remains one of the most lethal cancers largely due to disseminated disease at the time of presentation. Here, we investigated the role and mechanism of action of the metastasis-associated protein anterior gradient 2 (AGR2) in the pathogenesis of pancreatic cancer. AGR2 was induced in all sporadic and familial pancreatic intraepithelial precursor lesions (PanIN), PDACs, circulating tumor cells, and metastases studied. Confocal microscopy and flow cytometric analyses indic… Show more

“…It should be noted that AGR2 expression can also be upregulated in PDAC, as other publications have suggested. 14,16 Previous reports have documented conflicting results regarding the prognostic value of AGR2 expression. In prostate cancer tissue, AGR2 downregulation correlated with increased Gleason grade, a histological grading system of prostate cancer, and increased cancer recurrence in Gleason grades 4 or 5.…”

“…AGR2 immunoreactivity was determined using an intensity score, graded on a scale from 0 to 3, and a proportional score, corresponding to the percentage of stained cells (0: no staining, 1:o20%; 2: 20-50%, and 3:450%), as previously described. 16 The total score was the product of intensity and the proportional score. AGR2 high-expressing patients scored between 4 and 6 (n ¼ 139) and low-expressing patients between 0 and 3 (n ¼ 54).…”

“…Agr2 is overexpressed in various cancer cells, and aberrant Agr2 expression in several tumor types predicts worse clinical outcomes (32)(33)(34)(35)(36)(37). Previously, however, the functions of secreted Agr2 and its significance in the progression of gastric SRCC had not been extensively examined.…”

“…AGR2 expression was shown to be significantly increased in HER2 positive breast tumors [147]. High levels of AGR2 were also associated with cell dissemination of pancreatic carcinoma [148] and with poor prognosis of lung cancer [149]. Worth of note, this protein appears highly represented in the tumor tissues (when present) and almost absent in the normal ones.…”
