Age-specific penetrance of LRRK2 G2019S in the Michael J. Fox Ashkenazi Jewish LRRK2 Consortium

Abstract: Objective: Estimates of the penetrance of LRRK2 G2019S vary widely (24%-100%), reflective of differences in ascertainment, age, sex, ethnic group, and genetic and environmental modifiers. Methods:The kin-cohort method was used to predict penetrance in 2,270 relatives of 474 Ashkenazi Jewish (AJ) Parkinson disease (PD) probands in the Michael J. Fox LRRK2 AJ Consortium in New York and Tel Aviv, Israel. Patients with PD were genotyped for the LRRK2 G2019S mutation and at least 7 founder GBA mutations. GBA mutati… Show more

“…A higher relative proportion of LRRK2 carriers among women (45 vs. 55%) vs. men (40 vs. 60%) was observed 25, 26. However, we did not demonstrate a difference between the rates of PD in male and female LRRK2 mutation carrier PD (52 vs. 48%).…”

“…Although genetic determinants may be sexually dimorphic,21, 22 and a specific LRRK2 gender effect has been postulated,23 we confirm that the male predominance observed in Western populations with PD is not present in the G2019S mutation in LRRK2 PD 23, 24, 25. A higher relative proportion of LRRK2 carriers among women (45 vs. 55%) vs. men (40 vs. 60%) was observed 25, 26.…”

“…Penetrance increases with age until 25%-75% at 80 years according to different studies. 2,19,20 As expected, PD converters in our study were older (an average of 10 years) than nonconverters, with statistical nonsignificance probably just because of the small number of cases. One of the patients was younger than 60 years when he developed PD motor symptoms.…”

“…7,8 Healthy carriers of the LRRK2 G2019S mutation therefore form a unique population at risk of PD. 9,10 Penetrance estimations range from 10%-17% at the age 50 years to 25%-85% at the age of 70 years, 7,[11][12][13] suggesting that the risk of developing PD is age-dependent. 7 Thus, we investigated age-related changes in corticostriatal connectivity in healthy LRRK2 G2019S mutation carriers.…”
