Age-related regional differences in cardiac nerve growth factor expression

Abstract: Age has been identified as an independent risk factor for cardiovascular diseases. A shift of the cardiac autonomic nervous system towards an increase in sympathetic tone has been reported in the elderly. Nerve growth factor (NGF) is the main neurotrophic factor that increases the sympathetic activity of the heart. If there is a shift of NGF expression in old compared to young cardiomyocytes and whether there are regional differences in the heart still remain unclear. Therefore, we chose a rat model of differe… Show more

“…A gradual increase in Ngf and TrkA expression also occurs with age. 29,30 These findings are consistent with a contribution of NGF to the change in autonomic tone observed in elderly patients. 30 At 60 days, NGF protein was significantly increased in the intact VP versus the sham despite the fact that Ngf RNA expression was highest in the denervated VP.…”

This increase in intact NGF protein suggests that with nerve perturbation, the adolescent, neurally intact prostate has both the appropriate neural signal and an undiminished capacity for epithelial response.…”

“…121 Interestingly, another study demonstrated an age-dependent increase of ET-1/NGF protein expression in both atria, indicating the possibility that sympathetic hyperinnervation induced agerelated occurrence of AF. 166 However, because AF is a complex disease with multiple possible mechanisms, including inflammation, oxidative stress, and so on, further investigation is warranted to elucidate the precise mechanisms at play under specific conditions.…”

“…Since the heart is comprehensively innervated by neural structures, including sympathetic and parasympathetic input, the influence of the local cardiac autonomic nervous system has to be considered in particular to draw a complete picture regarding cardiovascular events. The knowledge that heart muscle cells age dependently control the autonomic nervous system due to autocrine / paracrine mechanisms including the expression and secretion of acetylcholine and nerve growth factor, leading to a sympathetic hyperinnervation in the elderly, underlines this assumption [ 44 , 45 ]. In regard to this, further attention has to be given to the fact, that the occurrence of TCM is not only gender dependent in most cases, TCM occurs predominantly in the elderly.…”
