Age-related changes in conversion of 5 -androstan-17 -ol-3-one to 5 -androstane-3 ,17 -diol and 5 -androstane-3 ,17 -diol by rat testicular cells in vitro

Abstract: Conversion of labelled 5 alpha-androstane-17 beta-ol-3-one (DHT) by isolated testicular cells from rats of different ages was examined under saturating substrate conditions in vitro (5--10 micrograms DHT/ml in a 24 h incubation). Two detectable metabolites of DHT were produced by testicular cells in vitro. 5 alpha-androstane-3 alpha, 17 beta-diol (3 alpha-diol) and 5 alpha-androstane-3 beta, 17 beta-diol (3 beta-diol). Production of these diols during a 24 h period was linear, and the amounts formed were direc… Show more

“…In addition, we attempted to describe the relationship between Leydig cell T production and peripheral androgen levels by measuring serum T concentrations in animals that were processed to obtain Leydig cells. In all cases, T concentrations were assayed in aliquots of spent media and serum samples by a previously described tritium-based RIA with an interassay variation of 7%-8% [48]. In order to localize BPA-induced lesions, we analyzed the levels of LHCGR, steroidogenic acute regulatory protein (STAR) and steroidogenic enzymes (CYP11A1, HSD3B, CYP17A1, and HSD17B3) in Western blots of ALCs isolated at 90 days of age from control and adult male rats exposed to BPA in the perinatal period (i.e., GD 12-PND 21).…”

“…Testosterone, androstenedione, and androstanediol were measured by RIA in plasma, in conditioned medium from in vitro LC cultures, and in supernatants of total testicular homogenates (Coffey et al 1971, Cochran et al 1979, 1981. All samples were assayed in duplicate, and each experimental data point consisted of 3-6 samples.…”

“…Testosterone, androstenedione, and 5␣-androstane-3␣-17␤-diol (androstanediol) rather than 5␣-androstane-3␤-17␤-diol were measured in conditioned media from in vitro cultures and supernatant of total testicular homogenates (22)(23)(24). All samples were assayed in duplicate.…”
