Age-Dependent Changes in Nrf2/Keap1 and Target Antioxidant Protein Expression Correlate to Lipoxidative Adducts, and Are Modulated by Dietary N-3 LCPUFA in the Hippocampus of Mice

et al.

Abstract: The brain has a high metabolism rate that may generate reactive oxygen and nitrogen species. Consequently, nerve cells require highly efficient antioxidant defenses in order to prevent a condition of deleterious oxidative stress. This is particularly relevant in the hippocampus, a highly complex cerebral area involved in processing superior cognitive functions. Most current evidence points to hippocampal oxidative damage as a causal effect for neurodegenerative disorders, especially Alzheimer’s disease. Nuclea… Show more

“…Estimation of membrane viscosity (in reciprocity to fluidity) from lipid profiles was performed following the statistical approaches reported for mouse brains [ 70 , 72 ]. A predictive multivariable function based on multiple regression analyses, which integrates the contributions of the phospholipid-to-cholesterol ratio, unsaturation-index-to-saturates ratio, average carbon chain length, and anionic-to-zwitterionic-phospholipids ratio were found to accurately predict membrane microviscosity (R 2 = 0.68, p < 0.005) [ 73 ].…”

“…The Nrf2 protein serves as a vital transcription factor essential for preserving the integrity of redox signaling when cells face oxidative stress [10,11]. As a member of the cap'n'collar basic leucine zipper transcription factor family, Nrf2 plays a vital role in coordinating antioxidant and detoxification responses by upregulating downstream genes [12][13][14]. Under normal conditions, Nrf2 predominantly resides in the cytoplasm, forming a complex with its inhibitory partner, Kelch-like ECH-associated protein 1 (Keap1).…”
