Age dependence of tissue plasminogen activator concentrations in plasma, as studied by an improved enzyme-linked immunosorbent assay.

Abstract: A procedure for improving the specificity of enzyme-linked immunosorbent assays (ELISA) was devised, based on addition of antigen-specific or non-immune immunoglobulins to the citrated plasma sample and defining the difference in assay response between these two mixtures as the antigen-specific part of the response. When applied to measurement of tissue plasminogen activator (t-PA; EC 3.4.21.31) antigen in plasma, this procedure resulted in elimination of the overestimates obtained in a large proportion (10-20… Show more

“…Plasma tissue plasminogen activator (tPA) antigen concentrations were determined in citrated plasma by an enzyme-linked immunosorbent assay (Imulyse. Biopool AB, UmeA, Sweden) [5]. Plasma plasminogen activator inhibitor (PAI) activity was measured in citrated plasma with a chromogen substrate assay (Spectrolyse/pL, Biopool AB, Umel, Sweden) [6].…”

“…EC-SOD in plasma was analysed by an enzyme-linked immunosorbent assay [28], as were the concentrations of tissue plasminogen activator, tPA [29]. tPA activity was determined with a parabolic rate assay based on fibrin stimulation of the tPA-mediated conversion of plasminogen and plasmin using the Spectrolyse/fibrin kit from Biopool AB (Umeå, Sweden) [27,30].…”
