AFLP genetic polymorphism in wild barley (Hordeum spontaneum) populations in Israel

Abstract: The genetic diversity produced by the amplified fragment length polymorphism (AFLP) method was studied in 94 genotypes of wild barley, Hordeum spontaneum (C. Koch) Thell., originating from ten ecologically and geographically different locations in Israel. Eight primer pairs produced 204 discernible loci of which 189 (93%) were polymorphic. Each genotype had a unique banding profile and the genetic similarity coefficient varied between 0.74 and 0.98. The phenogram generated from these similarities by the UPGMA … Show more

“…Most of the differentiation (95%) was present within accessions. This could be due to the natural selection and gene flow as it had been reported in many studies (Turpeinen et al, 2003;Nevo et al, 2004;Shakhatreh et al, 2015). According to Nevo et al (1998), natural selection appears to be a major differentiating and orienting force of regional evolutionary change, maintaining genetic polymorphisms under conditions of environmental heterogeneity and stress.…”

“…This species has been intensively studied for the extent and structure of genetic diversity at a wide range of spatial scales from tens to hundreds and even thousands of kilometers (Brown et al, 1978a;Turpeinen et al, 2001Turpeinen et al, , 2003Volis et al, 2001bVolis et al, , 2002aVolis et al, , 2003Volis et al, , 2005Morrell et al, 2003). At large geographical scale, selection and colonization history are predominant evolutionary forces, and a great deal of information about these processes has been gained for wild barley.…”

“…Varied patterns of polymorphism and geographic structure are reported at different wild barley loci (12,13). Selective association between genetic diversity in wild barley and ecogeographical factors has been shown by using applied fragment length polymorphism (14,15) and microsatellite (16,17) markers. We report here a study of diversity in the coding region of the Isa gene at the DNA sequence level.…”
