AFLP-Based Genetic Linkage Map for the Red Flour Beetle (Tribolium castaneum)

Abstract: The red flour beetle (Tribolium castaneum) is a major pest of stored grain and grain products and a popular model species for a variety of ecological, evolutionary, and developmental biology studies. Development of a linkage map based on reproducible and highly polymorphic molecular markers would greatly facilitate research in these disciplines. We have developed a genetic linkage map using 269 amplified fragment length polymorphism (AFLP) markers. Ten previously known random amplified polymorphic DNA (RAPD) m… Show more

“…Previously described markers (Zhong et al 2004;Demuth et al 2007) were used to screen for polymorphism between cSM and TIW1 parent strains. Of the 42 markers previously reported on LG3, 9 were polymorphic between the two parental strains.…”

“…The sequences of forward and reverse primers are listed in Supporting Information, Table S1. Marker polymorphism was screened using the method described previously (Zhong et al 2004 and the Li-Cor model 4300 automated DNA analyzer (Li-Cor, Lincoln, NE). The Gene ImagIR 4.33 software (LiCor) program was used to quantify allele size based on the pattern of the height of signal peaks.…”

“…AFLP analysis is a DNA fingerprinting technique that gives several hundred fragments in a single PCR reaction, and it is possible that some strain-specific fragments may be identified (Rieseberg et al 1999). AFLP markers have been widely used in genetic diversity and genetic mapping studies and are useful for studying genetic introgression in wild populations (Maughan et al1996;Zhu et al 1998;Yan et al 1999;Zhong et al 2004). AFLP's dominant nature is a disadvantage because each marker is represented by only two alleles, one of them being unamplifiable (null), thereby decreasing their accuracy in allele frequency estimation in comparison with codominant markers (Lynch and Milligan 1994).…”
