AFLP and SCAR markers linked to the suppressor gene (Rf) of a dominant genetic male sterility in rapeseed (Brassica napus L.)

Abstract: Rs1046AB is a line which is true breeding for a dominant genetic male sterility gene (Ms) but which is a mixture of male fertile and sterile individuals (a two-type line) because it is segregating for a dominant suppressor gene (Rf). This system provides a promising alternative to the CMS system for hybrid breeding in Brassica napus. In order to identify molecular markers linked to the Rf gene, a near-isogenic line (NIL) population from the cross between a sterile individual (MsMsrfrf) and a fertile individual… Show more

“…AFLP can produce large number of candidate markers in a single experiment and it is highly reproducible [ 33 ]. However, AFLP markers are relatively costly and the method is technologically demanding, and this limits its application in breeding programs [ 34 ]. Thus, we converted an AFLP marker into a convenient and inexpensive PCR-based marker SCAR.…”

“…Linkage analysis showed that three markers, designed as SCHDA, SCHDC and SCHDF, were co‐segregating with each other. They all had the same genetic distance of 0.7 cM from the Rf gene (Hong et al. 2006).…”

“…2004a,b). Recently, the Rf gene was also tagged with six AFLP polymorphic fragments and the three nearest were converted to SCAR markers (Hong et al. 2006).…”

“…Crossing dominant GMS plants with recessive temporary maintainer lines can produce all male sterile progenies as female parents for hybrid seed production. Numerous DNA markers closely linked with GMS genes have been developed for marker-assisted breeding and have contributed to positional cloning (Ying et al 2003;Ke et al 2004;Lu et al 2004;Ke et al 2005;Hong et al 2006;Song et al 2006;He et al 2008;Hong et al 2008;Feng et al 2009;Xu et al 2009). A total of five GMS genes were mapped on linkage groups N7, N16, N19 and A8 in B. napus (Yi et al 2006;Huang et al 2007;Lei et al 2007;Xiao et al 2008;Lu et al 2013).…”
