Affinity-Switchable Lateral Flow Assay

Abstract: Lateral flow assay (LFA) has been a valuable diagnostic tool in many important fields where rapid, simple, and on-site detection is required, for applications such as pregnancy tests and infectious disease prevention. Currently, two types of LFAs are available: lateral flow immunoassay (LFIA) and nucleic acid lateral flow assay (NALFA). Both are generally used for the testing of proteins and nucleic acids. However, enzyme activities and small molecules without the corresponding binding partner cannot be detect… Show more

“…Apart from the hybridization reaction and antibody–antigen interaction, the avidin-biotin system has been extensively used in biomedical analysis. Avidin and its analogues, including streptavidin (SA) or neutravidin (NA), modified on a solid surface show high stability, easy controllability and excellent flexibility [ 35 , 36 , 37 ]. Many SA or NA-labeled signal tags or solid supports and biotinylated reagents are commercially available.…”

“…In order to develop the improved sensing strategies, gold NPs were prepared in advance and subsequently offered sensing responses in solid phases, such as woods and hydrogels. [23][24][25][26] Hydrogel is a useful environment-friendly material, which has a polymer framework structure. 27,28 The hydrogels have been widely used for improvements in biotechnology and nanotechnology.…”

“…Traditional colorimetric ELISAs and modern chemiluminescence ELISAs need sophisticated instruments or well-trained laboratory employees to analyze the results. In this regard, point-of-care (POC) diagnostics appear to be very suitable for on-site patient care and long-term disease monitoring in a facile manner. Immunochromatography test strip (ICTS), the most widely used type of POC, has been recently developed for the qualitative, semiquantitative, and even quantitative detection of serum biomarkers, viruses, toxins, bacteria, and proteins. …”
