Aerosol Delivery of Liposomal Formulated Paclitaxel and Vitamin E Analog Reduces Murine Mammary Tumor Burden and Metastases

Abstract: Paclitaxel is a chemotherapeutic agent used for the treatment of metastatic breast cancer. 2,5,7,8-Tetramethyl-2R-(4R, 8R-12-trimethyltridecyl) chroman-6-yloxyacetic acid (alpha-TEA) is an analog of vitamin E that inhibits primary tumor growth and the incidence of lymphatic and pulmonary metastases in preclinical animal models. Here, the efficacy of sequential treatment with paclitaxel and alpha-TEA was tested in the BALB/c syngeneic 66cl-4-GFP mammary cancer model. Both agents were formulated into liposomes a… Show more

“…Synergetic anti-cancer effect was observed even if platinum-resistant tumor was used [83]. The developed aerosol formulation of liposomal α-TEA was successfully combined with liposomal 9-nitro-camptohecin, liposomal paclitaxel or celecoxib to reduce tumor burden as well as lung and lymph node metastases in various animal cancer models [84][85][86][87].…”

“…Encouragingly, in a recent phase I study, liposomal formulations of cisplatin have successfully been delivered to the lungs via aerosol delivery with higher drug deposition at the tumour site and minimal systemic toxicity [51]. The pulmonary delivery of liposomal 9-nitrocamptothecin, doxorubicin and paxlitaxel has been explored in similar approaches [52][53][54].…”

“…As a negative control, parallel sections were incubated with nonimmune rabbit IgG 1 sera (prediluted; Dako North America). Slides were also stained with anti-Ki67 to determine the fraction of S-phase nuclei and anti-CD31 antibody (BD Pharmingen, San Diego, CA) to quantify vascular density and apoptotic nuclei were detect by TUNEL assay (Latimer et al, 2009).…”
