Background: Chemotherapy accounts to high vulnerability toward Adverse Events (AEs) as it involves highly complicated regimens. Objective of the study is to identify, assess, grade and categorize chemotherapy-induced adverse drug reactions in pediatric oncology. Materials and Methods: A 6 months Prospective-observational study was conducted in pediatric hematology oncology department. 0-15 years of hospitalized patients receiving chemotherapy for cancer were enrolled. Demographic details, cytotoxic drugs used and AEs that occurred were collected in a self-designed patient profile form. AEs were studied using CTCAE version 4.03 and Naranjo's Scale of Probability. After each cycle, follow ups were taken. Result: A total of 50 patients, of which 33 male and 17 female patients were included in the study and a total of 332 AEs were observed. AEs were segregated into organ class and the number of events observed were, Gastrointestinal (123), General Disorders (107), Musculoskeletal (23) be caused by more than one chemotherapeutic drug. Using Naranjo's Scale we observed, 6 were definite, 159 were probable and 167 were possible. On using CTCAE, out of 332 AEs, 167 were characterized as Grade 1, 114 as Grade 2, 51 as Grade 3 and 1 as Grade 5. Conclusion: Using CTCAE, most AEs were suspected to have been caused because of Daunorubicin and Methotrexate, but the severe AEs (neurotoxicity, pulmonary toxicity) were due to Vincristine, Methotrexate, and Cyclophosphamide.

Aims Pharmaco‐epidemiological surveys enable the frequency of serious adverse effects—and also the determining factors of their occurrence and seriousness—to be quantified. Few studies systematically gathering post‐chemotherapy adverse effects data have been conducted. The objective was to assess the incidence of post‐chemotherapy serious adverse effects on the basis of cancer registry data. Methods The population was composed of new invasive cancer cases, with the exception of haematopoietic tumours and cutaneous carcinomas. These cancers were identified in 2012 among patients living at the time of diagnosis in a region covered by a general cancer registry and by a French regional pharmacovigilance centre, and treated with neo‐adjuvant and/or adjuvant first‐intention chemotherapy, followed or not by radiotherapy. The study was based on a sample of 1000 patients from the registry, followed by the collection of serious adverse effects and the required information to constitute a pharmacovigilance file. Results Chemotherapy was associated with a particularly high incidence of serious adverse effects, affecting 44.5% (41.4–47.5%) of the patients. The highest incidence rates were observed when patients were exposed to topo‐isomerase II inhibitors such as etoposide and bleomycin (69.2%), vinca‐alkaloids (66.7%), topo‐isomerase I inhibitors (54.5%) and platinum derivatives (52.0%). The clinical context was also linked to incidence, especially in case of metastases (53.3%) and comorbidities (51.3%). Substantial differences were found according to localisation, with a particularly high incidence in bronchial–pulmonary cancers (59.0%). Conclusion The high overall incidence rate of serious adverse effects should motivate a reinforcement of information about drug toxicities and improve knowledge by drawing on patient reporting.

Cancer treatments usually cause adverse drug reactions. Therefore, safe anticancer drugs are needed in the treatment of cancer. One source of medicine that can be explored is plant. Extracts of longan leaves (Dimocarpus longan), jamaican cherry leaves (Muntingia calabura), and avocado leaves (Persea americana) have been tested for cytotoxic activity against several cancer cell lines. This study aims to determine the cytotoxic activity of ethanolic extract of longan leaves, jamaican cherry leaves, and avocado leaves against T47D and WiDr cells and to identify secondary metabolites in the extracts which have the highest activity. Ethanolic extract of longan leaves, jamaican cherry leaves, and avocado leaves were tested for their cytotoxic activity using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method. Identification of secondary metabolites in the ethanolic extract of avocado leaves was carried out by thin layer chromatography method using silica gel GF254 as the stationary phase and a mixture of n-hexane and acetone (6:4) as the mobile phase. Cytotoxic test results show that ethanolic extract of longan leaves and cherry leaves with concentration of up to 1600 μg/mL do not reduce the T47D and WiDr living cells to 50%. Avocado leaf extract decreases the percentage of living T47D cells and WiDr with IC50 values of 790.679 µg/mL and 1072.2 µg/mL, respectively. The ethanolic extract of avocado leaves contains flavonoid, phenolic, and terpenoid. Ethanolic extract of longan leaves, cherry leaves and avocado leaves do not have cytotoxic activity against T47D and WiDr cells.
