Advantages in Wound Healing Process in Female Mice Require Upregulation A2A-Mediated Angiogenesis under the Stimulation of 17β-Estradiol

Abstract: Estrogenic steroids and adenosine A2A receptors promote the wound healing and angiogenesis processes. However, so far, it is unclear whether estrogen may regulate the expression and pro-angiogenic activity of A2A receptors. Using in vivo analyses, we showed that female wild type (WT) mice have a more rapid wound healing process than female or male A2A-deficient mice (A2AKO) mice. We also found that pulmonary endothelial cells (mPEC) isolated from female WT mice showed higher expression of A2A receptor than mPE… Show more

“…Adenosine receptors A2a and A2b are prevalent on endothelial cells and mediate the adenosine response to injury through cAMP/MAPK/ERK pathway signaling [ 29 ]. These receptors are specifically regulated by both hypoxia and estrogen, however, crosstalk between adenosine generation and estrogen remains poorly understood [ 30 , 31 , 32 ]. Our data demonstrate the reduction in adenosine with the inhibition of estrogen receptors and conversely, with estradiol exposure, decreased extracellular ATP.…”

“…Pups were sexed by visual evaluation and confirmed by PCR using the expression of the jarid gene, as previously described. 28 Then, the weight-for-length ratio and weight-for-weight ratio of the brain were calculated individually. After that, brains were collected in 300 ml of RIPA buffer and frozen at À20 C until further analysis.…”

“…The in vivo wound-healing assay was performed as previously described by our laboratory [ 36 , 37 ], with slight modifications. Briefly, WT and Thy-1 −/− male mice were maintained separately in individual cages.…”
