Objective — To investigate the apoptotic activity of Gratiola officinalis L. extract on human tumor cell lines by flow cytofluorometry. Material and Methods — The extract of Gratiola officinalis L. was manufactured via our original methodology. Studies were performed on human tumor cell lines: HeLa – cervical carcinoma, Jurkat – T-cell lymphoblastic leukemia, MCF-7 – breast adenocarcinoma, A549 – lung carcinoma, PC-3 – prostate carcinoma, HCT-116 – colon carcinoma, A498 – renal carcinoma, and SK-BR-3 – human breast carcinoma. Induction of apoptosis was studied after incubating cell lines with Gratiola officinalis L. extract at a concentration of 0.9 mg/mL using the Annexin V-FITC Apoptosis Kit. Caspase-dependent apoptosis was examined on a flow cytometer using anti-caspase-3-FITC (BD) kit on the Jurkat cell line. Morphological studies of HeLa cervical carcinoma cells in the alive and dead test were performed using two stains, acridine orange and propidium iodide, at different concentrations of the extract. The statistical data processing was performed using Microsoft Office Excel software. Results — One day after their exposure to Gratiola officinalis L. extract at a concentration of 0.9 mg/mL, tumor cells were mostly in late apoptosis stage. Cytotoxic activity of Gratiola officinalis L. extract was established for all investigated tumor cell cultures but their sensitivities to the extract were different. Mechanisms of antitumor action of Gratiola officinalis L. extract were identified: we established that the extract induced caspase-dependent apoptosis in tumor cells. Conclusion — The identified mechanisms of apoptotic activity of Gratiola officinalis L. extract confirmed the prospects of bioflavonoids as new-generation antitumor agents.

We compared the effectiveness of antitumor effects and apoptosis induction on human renal cell carcinoma A498 extracts of cruciferous plant material (indole-3-carbinol), Chinese mushroom (cordycepin), French red wine (resveratrol) at low concentrations after 24 and 48 h using fluorescent methods of imaging apoptosis and necrosis in human tumor cells in vitro . Propidium iodide and acridine orange were used as dyes in the "alive and dead" test, which allowed us to detect the number of dead cells and cells in which apoptosis had started. It was found that indole-3-carbinol at low concentrations (0.0288 and 0.1152 mg/ml) has a pronounced cytotoxic and cytostatic activity against human kidney cancer cells, significantly exceeding the action of resveratrol at the same concentrations. At the same time, cordycepin has no cytotoxic and cytostatic activity at these concentrations. Indole-3-carbinol also has the most pronounced apoptotic activity at concentrations of 0.0144-0.1152 mg/ml, after 48 h the number of kidney cancer cells in apoptosis increases by 6.8-10 times compared to control. It is concluded that indole-3-carbinol is a promising antitumor agent for use in the complex therapy of patients with kidney cancer.

С использованием флуоресцентных методов визуализации апоптоза и некроза в опухолевых клетках человека in vitro сравнивались эффективности противоопухолевого воздействия на клетки карциномы почки человека А498 флавоноидсодержащих экстрактов: аврана лекарственного и зеленого чая в сверхнизких концентрациях через 24 и 48 h. Флуоресцентные красители (йодистый пропидий и акридиновый оранжевый) использовались в тесте " живые и мертвые", что позволило выявлять общее количество погибших клеток путем некроза и апоптоза, а также клеток, в которых запустился апоптоз, при этом выявлялись апоптотические тельца или пикноз ядра. Наиболее выраженной цитостатической активностью через 48 h после воздействия обладал экстракт аврана, уменьшая количество клеток в поле зрения в 1.5 раза по сравнению с контролем. Наиболее выраженной цитотоксической активностью обладал экстракт зеленого чая в концентрации 0.0288 mg/ml, и выраженность ее нарастала через 48 h после воздействия. Экстракт аврана проявлял цитотоксическую активность при более низких концентрациях (0.0036 mg/ml).Ключевые слова: флуоресцентные методы визуализации, противоопухолевая активность, клеточная культура, цитостатическая активность, флавоноиды, рак почки, авран лекарственный.
