MicroRNAs control cellular processes by regulating expression of their target genes. Here we report that neuro-epithelial transforming gene 1 (NET1) is a target of tumor suppressor microRNA 22 (miR-22). miR-22 is downregulated in peripheral blood mononuclear cells derived from chronic myeloid leukemia (CML) patients and in CML cell line K562. NET1 was identified as one of the targets of miR-22 using both in vitro and in vivo experiments. Either mutations or naturally occurring single-nucleotide polymorphisms in NET1 3 0 -UTR that map at the miR-22 binding site were found to affect binding of miR-22 to NET1 mRNA. Over expression of NET1 in K562 cells resulted in increased proliferation. However decreased proliferation and alteration in cell cycle were observed on either overexpression of miR-22 or knockdown of NET1 expression respectively. We also found that overexpression of miR-22 or NET1 knockdown inhibits actin fiber formation, probably by downregulation of NET1 as NET1 knockdown also resulted in depletion of actin fiber formation. We suggest that the oncogenic properties of CML cells are probably due to deregulated expression of NET1 as a result of altered expression of miR-22.

Micro RNA (miRNA) is a non-coding RNA (ncRNA) which was discovered in 1993. It consists of 19-25 nucleotides and it regulates the expression of around 30% of protein coding mRNAs in humans. Recent studies on miRNAs suggest the fact that it has a vital role in the post-transcriptional gene regulation of skin development and in future it can also be used for treating skin diseases. Certain number of novel miRNAs has been identified in skin and various studies were done on the functional roles of miRNAs in psoriasis. In this manuscript, we review the profiling and characterization of miRNAs in the psoriatic skin of humans and discuss its implication in various biological activities and we also share our views on the application of Bioinformatics in predicting novel miRNAs along with the nanotechnology based miRNA delivery to understand the disease pathology of Psoriasis which is followed by treatment. There is a broader scope for addressing the challenges in miRNA based drug delivery in future.

Co- and post-transcriptional regulation of gene expression is complex and multi-faceted, spanning the complete RNA lifecycle from genesis to decay. High-throughput profiling of the constituent events and processes is achieved through a range of technologies that continue to expand and evolve. Fully leveraging the resulting data is non-trivial, and requires the use of computational methods and tools carefully crafted for specific data sources and often intended to probe particular biological processes. Drawing upon databases of information pre-compiled by other researchers can further elevate analyses. Within this review, we describe the major co- and post-transcriptional events in the RNA lifecycle that are amenable to high-throughput profiling. We place specific emphasis on the analysis of the resulting data, in particular the computational tools and resources available, as well as looking towards future challenges that remain to be addressed.
