Pesq. Vet. Bras. 34(9):874-884, setembro 2014 874 RESUMO.-O mastocitoma é a neoplasia cutânea maligna de maior frequência em cães. Apesar do estudo e do desenvolvimento de diversos marcadores prognósticos na tentativa de prever o comportamento biológico deste tipo de neoplasia, a graduação histológica continua sendo utilizada como principal delineador prognóstico para os animais acometidos, mesmo não conseguindo prever o comportamento de mastocitomas de diferenciação intermediária. Diferentes marcadores de proliferação e atividade celular vêm sendo testados com resultados promissores, entretanto, em sua maioria, demandam técnicas específicas de coloração e imuno-histoquímica que, ainda hoje, são de alto custo e muitas vezes não estão facilmente disponíveis. O índice mitótico é um método indireto de mensuração da atividade proliferativa celular, que pode ser contabilizado em uma lâ-mina comum de avaliação histológica e estudos vêm determinando-o como importante marcador prognóstico para o mastocitoma canino. Dada a recente publicação sugerindo nova graduação em dois graus (alto e baixo) e indisponibilidade das técnicas específicas utilizadas na determinação (MCT) is the most common malignant cutaneous neoplasm in dogs. Even with several prognostic markers being studied, in attempt to previse the biological behavior of these tumors, the histological grading is still commonly used to predict behavior, however, it is not significant to predict intermediately differentiated MCT behavior. Different prognostic and cell proliferation markers are being tested with promising results however, the majority of then requires specific staining and immunohistochemical techniques that even nowadays are expensive and not suitable for all veterinarians. Mitotic index is an indirect measure of cell proliferation and it can be established in a common histological microscope slide, and studies indicate it as an important prognostic marker for MCT in dogs. Given the recent publication of the novel two-tier histologic grading system, this study objective to evaluate the prognostic value of the different grading systems associating with survival and compare it with the mitotic index. Even though both grading systems associate with survival in the univariable analyses (P<.001), at the multivariable analyses neither was associated with survival (P>0,05), and the mitotic index was the most significant prognostic factor for survival, regardless of the cut-off point used (P<0,05), especially when Patnaik grade 2 tumors with unpredictable biological behavior were analyzed. The mitotic index was also significantly associated with both local and distant reccurence and paraneoplastic syndrome. In this study, mitotic index was the most significant prognostic factor, in the absence of specific staining and advanced immunohistochemical techniques, and its use is indicated in association with both histological grading system, aiming to add higher prognostic value possible.INDEX TERMS: Cutaneous mast cell tumor, prognostic markers, mitotic inde...

In spite of the many available protocols, the use of chemotherapy for the management of canine mast cell tumours (MCT) remains empirical, and there is lack of criteria for the choice of protocol and definition of patients who may benefit from treatment. The objective of this study was to evaluate the outcome of dogs with MCT after adjuvant chemotherapy according to the risk of recurrence or metastasis proposed on the literature. This prospective study included 89 followed up dogs with prognosis assesment including clinical, histological, immunohistochemical and genetic features of canine MCT. Patients were grouped according to risk of recurrence and metastasis and recommended treatment with lomustine followed by chlorambucil if considered at high-risk, or vinblastine followed by chlorambucil if a patient was at intermediate risk. Outcome was defined by disease-free interval (DFI) and overall survival (OS) estimated by Kaplan-Meier curve. Adjuvant lomustine was useful for control of canine MCT of high-risk of recurrence or metastasis, but only when sequentially associated to chlorambucil with a DFI of 686 days and not reached OS.

RESUMO.O mastocitoma cutâneo é uma neoplasia observada frequentemente em cães. Esta doença é caracterizada pelo crescimento anormal e excessivo de mastócitos. Este estudo realizou o levantamento de dados de todos os arquivos de biopsia feito em cães no laboratório de Patologia clínica do Centro Universitário da Grande Dourados e da Universidade Anhaguera Uniderp no período entre janeiro de 2015 e janeiro de 2016. Foram selecionados 26 casos confirmados de mastocitoma canino, a partir da informação retiradas dos laudos foram avaliados fatores predisponentes como raça, sexo, idade e local das lesões neoplásicas. Em meio aos animais selecionados 46,2% (12) eram fêmeas e 53,8% (14) machos. Com relação ao fator racial, 34,6% (9) dos animais não possuem raça definida, 30,8% (8) eram da raça Boxer, 11,5% (3) eram da raça Pitbull, 7,7% (2) da raça Labrador, 7,7% (2) eram da raça Dachshund, 3,8% (1) era da raça Poodle e 3,8% (1) da raça Shnauzer. Concluiu se neste trabalho que a maioria dos cães acometidos por essa neoplasia eram sem raça definida. Quando considerado a raça observou se que os cães Boxer são os mais predispostos a esta neoplasia. Animais considerados idosos, com idade entre 9 e 13 anos são os mais acometidos. O mastocitoma canino não apresentou predisposição à doença quanto ao sexo e pôde-se verificar a maior frequência em relação à localização dos nódulos na parte do tronco e membros.Palavras chave: Mastócitos, células redondas, tumores. This disease is characterized by abnormal and excessive growth of mast cells. This study carried out the data collection of all biopsy files made in dogs in the clinical pathology laboratory of the University Center of Grande Dourados and Universidade Anhaguera Uniderp between January 2015 and January 2016. Twenty-six confirmed cases of mastocytoma From the information collected from the reports, predisposing factors such as race, sex, age and site of neoplastic lesions were evaluated.
