Drought represents the most significant environmental challenge impacting the sugar beet growth and productivity. Sodium azide (NaN3) is relatively safe to handle and the most efficient mutagen. So, the objective of the present investigation was to study the effect of presoaking two sugar beet cultivars seeds in various concentrations of sodium azide to improve drought tolerance in vitro. Results showed that the polyethylene glycol (PEG) at 5% or 10% caused water stress in medium and lead to highly significant reduction in shoot length, fresh weight, leaves number and photosynthetic pigment (Chl a, b and car). The 10% of PEG-6000 recorded the maximum reduction in growth parameter of two studied sugar beet cultivars. Otherwise, the tested sodium azide treatments improved sugar beet growth under stressed and unstressed plants. Presoaking of sugar beet seeds in 8 mM of sodium azide raises the shoot length, fresh weight, number of leaves and photosynthetic pigment of two cultivars and the most raising reported at unstressed plants. Proline content and antioxidant enzymes (POD, SOD and CAT) increased with increasing in poly ethylene glycol concentration. Moreover, sodium azide triggers the highest production of antioxidants enzymes in two cultivars of sugar beet under drought stress. Finally, presoaking of seeds in 4 or 8 mM of sodium azide can be enhancing drought tolerance in sugar beet.

Sugar beet is one of the greatest sources for producing sugar worldwide. However, a group of bacteria grows on beets during the storage process, leading to a reduction in sucrose yield. Our study focused on identifying common bacterial species that grow on beets during manufacturing and contribute to sucrose loss. The ultimate goal was to find a potential antibacterial agent from various plant extracts and oils to inhibit the growth of these harmful bacteria and reduce sucrose losses. The screening of bacterial species that grow on beet revealed that a large group of mesophilic bacteria, such as Bacillus subtilis, Leuconostoc mesenteroides, Pseudomonas fluorescens, Escherichia coli, Acinetobacter baumannii, Staphylococcus xylosus, Enterobacter amnigenus, and Aeromonas species, in addition to a dominant thermophilic species called Bacillus thermophilus, were found to be present during the manufacturing of beets. The application of 20 plant extracts and 13 different oils indicated that the extracts of Geranium gruinum, Datura stramonium, and Mentha spicata were the best antibacterials to reduce the growth of B. thermophilus with inhibition zones equal to 40, 39, and 35 mm, respectively. In contrast, the best active oils for inhibiting the growth of B. thermophilus were Mentha spicata and Ocimum bacilicum, with an inhibitory effect of 50 and 45 mm, respectively. RAPD-PCR with different primers indicated that treating sugar juice with the most effective oils against bacteria resulted in new recombinant microorganisms, confirming their roles as strong antibacterial products. The characterization of Mentha spicata and Ocimum bacilicum oils using GC/MS analysis identified cis-iso pulegone and hexadecanoic acid as the two main bioactive compounds with potential antibacterial activity. An analysis of five genes using DD-PCR that have been affected due to antibacterial activity from the highly effective oil from Mentha spicata concluded that all belonged to the family of protein defense. Our findings indicate that the application of these pure antibacterial plant extracts and oils would minimize the reduction of sucrose during sugar production.

Plant regeneration and dedifferentiation are known to be difficult for sugar beet (Beta vulgaris L.) due to its recalcitrant nature. This leads to low transformation efficiency and thus successful application of plant molecular techniques is limited in sugar beet for its genetic enhancement. A prolific regeneration method has been established by modulating several plant growth regulators on the in vitro regeneration of Beta vulgaris var. V6 KWS Serenada. Several types of explants excised from young seedlings of this variety were used for both direct and indirect regeneration of shoots. The highest response towards direct shoot formation and callus induction were obtained from cotyledonary nodes and hypocotyls, respectively. Explants were cultured on MS medium supplemented with different concentrations of BAP, GA3 and 2, 4 -D for callus induction as well as formation of shoots. Hypocotyls responded well for callus induction on MS medium containing 0.1 mg/l BAP + 2.0 mg/l 2, 4-D, while cotyledonary nodes exhibited the highest responses towards shoot formation on MS medium containing 1.0 mg/l BAP and 1.5 mg/l GA3. MS medium containing 2.0 mg/l IBA produced the highest number of roots per shoot. The in vitro raised rooted plantlets were successfully transferred to soil for acclimatization.
