Advancement in understanding the role of ferroptosis in rheumatoid arthritis

Abstract: Rheumatoid arthritis (RA) is a chronic, systemic disease of unknown etiology. The primary manifestation of RA is inflammatory synovitis, which eventually leads to deformity and functional loss. Ferroptosis is a non-apoptosis form of cell death that depends on intracellular iron accumulation. This leads to an increase in reactive oxygen species (ROS) induced-lipid peroxidation. The underlying mechanisms of ferroptosis are System Xc- and Glutathione metabolism, regulation of glutathione peroxidase 4 activity, an… Show more

“…In this study, twelve FRGs were differentially expressed between the synovium of RA patients and HCs. These genes participated in key mechanisms of ferroptosis, including GSH metabolism, iron metabolism and lipid metabolism [ 29 ]. Meanwhile, there were apparent differences in the correlations between FRGs in RA patients and HCs.…”

“…GPX4 relies on GSH to decrease lipid peroxide. As a critical regulator of ferroptosis, GPX4 is the most potent anti-lipid peroxidase in the cell ( 29 , 30 ). In order to produce GSH, System Xc − is required to exchange extracellular cystine and intracellular glutamate.…”

“…On the other hand, FLSs produce MMP to digest a variety of proteins in cartilage and supporting structures, further promoting the invasion and expansion of pannus ( 108 ). In addition, VEGF secreted by RA-FLSs is an important factor in promoting angiogenesis ( 109 , 110 ). Thus, it is evident that inducing ferroptosis of RA-FLSs not only regulates synovial homeostasis but also has important implications for the inhibition of angiogenesis.…”
