Advancement in protocol for in vitro seed germination, plant regeneration and cryopreservation of Viola cornuta

Abstract: The aim of this study was to develop a fast, reliable and true-to-type protocol for in vitro plant regeneration and long-term storage of horned pansy (Viola cornuta L). Seed germination over 60% was recorded after 12 weeks of growth at 10 °C or 4 °C. Calli formation and shoot induction were obtained in petiole and hypocotyl culture on half-strength MS mineral salts with full concentration of Na-FeEDTA and vitamins (½MS medium) with 2,4-dichlorophenoxyacetic acid (2,4-D, 0.1 mg/L) and 6-benzylaminopurine (BAP, … Show more

“…While hygromycin is indeed a selectable agent, cefotaxime may also affect the regeneration and growth of plant tissues in culture [80]. High percentages of callus formation and shoot regeneration (100% and 32%) from control explants indicate that the previously developed regeneration protocol from hypocotyl explants [56] is well optimized for V. cornuta cv. "Lutea Splendens".…”

“…"Lutea Splendens" were purchased from B and T World Seeds (Paguignan, France), case number 24421. They were surface sterilized and germinated as in [56]. The hypocotyl explants from upper part of in vitro germinated seedlings, close to epicotyl, were used as starting plant material for genetic transformation of V. cornuta.…”
