Adult Mammalian Neural Stem Cells and Neurogenesis: Five Decades Later

Abstract: Summary Adult somatic stem cells in various organs maintain homeostatic tissue regeneration and enhance plasticity. Since its initial discovery five decades ago, investigations of adult neurogenesis and neural stem cells have led to an established and expanding field that has significantly influenced many facets of neuroscience, developmental biology and regenerative medicine. Here we review recent progress and focus on questions related to adult mammalian neural stem cells that also apply to other somatic ste… Show more

“…The adult brain harbors germinal cell niches in the subventricular zone of the lateral ventricles and the subgranular zone in the dentate gyrus of the hippocampus (1)(2)(3). The activation of these relatively quiescent neural progenitor cell (NPC) 2 populations and their capacity to differentiate into specialized cells is under rigorous cellular control (4).…”

“…1), with the different markers, alone or in combination, designating the different phases, varying from quiescent stem cell states to committed precursor steps (reviewed in Vankelecom 2010, andChen 2014). Such heterogeneity within the stem cell population has also increasingly been demonstrated in other organs (Bond et al 2015;Donati and Watt 2015). Single-cell transcriptomic analysis of brain stem cells identified the presence of four subgroups in different stages of activation (most prominently in response to injury), ranging from dormant to primed-quiescent, active and active-dividing stem cells (Llorens-Bobadilla et al 2015).…”

“…The more extended neurogenic area in the adult mammalian brain is the V-SVZ, located along the entire length of the walls of the lateral ventricles (figure 1a; [6]). Formerly simply known as SVZ, its current denomination as V-SVZ has integrated the concept of the contribution of the ependymal cells (ECs) contacting the ventricle with the physiology of the neurogenic niche.…”
