Adult bone-marrow-derived mesenchymal stem cells contribute to wound healing of skin appendages

Abstract: Adult bone-marrow-derived mesenchymal stem cells (MSCs) are well-established as having the capacity to differentiate into cells with mesodermal, ectodermal, and endodermal characteristics and can leave their niche to home toward and engraft within foreign tissues. To investigate whether adult MSCs contribute to the repair of skin appendages after injury, BrdU-labeled MSCs were cocultured with heat-shocked confluent sweat gland cells (SGCs) in vitro and later intravenously injected into fullthickness skin wound… Show more

“…After centrifugation, the cells were re-suspended and counted with trypan blue stain 0.4%(100μL of cell suspension with 100μL of stain) using Neubauer haemocytometer as described by Belsey et al,1980 MSCs were distinguished from other BM cells by their tendency to adhere to tissue culture plastic (Mok et al,2003), and by negative expression of CD34 and positive expression of CD44 (Fig. 1) in immune-staining, described by Li et al,2006. Briefly, the cultured cells were fixed in petri dishes on day 12 of culture by Acetone: Methanol (1:1) then covered by H2O2 (10%).…”
