Spontaneous contractile activity of the circular smooth muscle of the guinea-pig seminal vesicle consists of twitch-like contractions with a mean frequency of 2.7/min.. The amplitude of responses to repetitive stimulation depended on the frequency of stimulation at constant pulse duration and on the pulse durations at constant frequency of stimulation. Mechanical summation in the tension development was observed at low frequency of repetitive stimulation. Adrenaline, noradrenaline and phenylephrine increased the frequency and amplitude of spontaneous contractions. The excitatory actions of adrenaline and noradrenaline were antagonized by phentolamine and phenoxybenzamine. DOPA; metanephrine and normetanephrine were not effective. Tyramine enhanced the spontaneous activity and its action was antagonized by phentolamine. Isoprenaline, at high concentrations, enhanced spontaneous activity, but in the presence of phentolamine, the spontaneous activity was inhibited by isoprenaline and orciprenaline. Isoprenaline inhibited the enhanced spontaneous activity induced by adrenaline, noradrenaline and tyramine. Spontaneous contractions were potentiated by tetrodotoxin, procaine, guanethidine and dibenamine but were not affected by nicotine and phenoxybenzamine. Evoked responses to repetitive stimulation were blocked or suppressed by tetrodotoxin, yohimbine, nicotine, guanethidine and phentolamine. The evoked responses were partially inhibited by phenoxybenzamine. In contrast, evoked responses were enhanced by hexamethonium and dibenamine. The role of the mucosa in generating the spontaneous activity of guinea-pig seminal vesicle (SV) was explored. Changes in contractility, membrane potential and intracellular Ca dynamics of SV smooth muscle cells (SMCs) were recorded using isometric tension recording, intracellular microelectrode recording and epi-fluorescence Ca imaging, respectively. Mucosa-intact but not mucosa-denuded SV preparations generated TTX- (1 μm) resistant spontaneous phasic contractions that were abolished by nifedipine (3 μm). Consistently, SMCs developed mucosa-dependent slow waves (SWs) that triggered action potentials and corresponding Ca flashes. Nifedipine (10 μm) abolished the action potentials and spontaneous contractions, while suppressing the SWs and Ca flashes. Both the residual SWs and spontaneous Ca transients were abolished by cyclopiazonic acid (CPA, 10 μm), a sarco-endoplasmic reticulum Ca -ATPase (SERCA) inhibitor. DIDS (300 μm) and niflumic acid (100 μm), blockers for Ca -activated Cl channels (CACCs), or low Cl solution also slowed or prevented the generation of SWs. In SV mucosal preparations detached from the muscle layer, a population of mucosal cells generated spontaneous Ca transients that were blocked by CPA but not nifedipine. These results suggested that spontaneous contractions and corresponding Ca flashes in SV SMCs arise from action potential generation due to the opening of L-type voltage-dependent Ca channels. Spontaneous Ca transients appear to primarily result from Ca release from sarco-endoplasmic reticulum Ca stores to activate CACCs to develop SWs. The mucosal cells firing spontaneous Ca transients may play a critical role in driving spontaneous activity of SV smooth muscle either by sending depolarizing signals or by releasing humoral substances.

The was deferens showed a phasic contraction followed by a tonic one in hyperosmotic 65 mM KC1 (hyper-65K+), isosmotic 60 mM or 120 mM KC1 solution. However, isosmotic 154 mM KC1 (iso-154K+) solution induced a large phasic contraction followed by a gradual relaxation. Applications of the high K+ solutions to the preparation of seminal vesicle showed almost similar results in tension to those of the was deferens. In the was deferens or seminal vesicle, an application of hyper-65K+ solution significantly decreased the relative value for the wet weight of the tissue at 120 min, but that of iso-154K+ solution did not affect the wet weight of both the muscles. In the was deferens, an hyperosmotic addition of sucrose at a concentration of 25 to 100 mM slightly prevented the tension decreased by the iso-154K+ solution and slightly decreased the relative wet weight. In the seminal vesicle, increasing sucrose concentration significantly increased the tension, but decreased the relative wet weight. In both the muscles of was deferens and seminal vesicle, an addition of pyrvate or oxalacetate (5.5 mM)to the iso-154K+ solution without glucose maintained the muscle tension at a maximal level in the iso-154K+ solution with glucose for 120 min. Further, the tension decrease in both the muscles induced by iso-154K+ solution was also prevented by hyperosmotic application of NaCl. From these results, it is suggested that the tension decrease by high K+, Na+deficient solution in the was deferens is mainly caused by an inhibition of glucose utilization by Nat-deficiency in the medium, however, the seminal vesicle is probably due to the cell swelling and partly the inhibition of glucose utilization by it.
