The present study was conducted to investigate the effects of the stems and leaves in ultrafined powder of Astragalus (SLASUP) on immune functions of chickens, and to provide a basis for clinical application of the stems and leaves of Astragalus. Two hundred and fifty Hyaline brown chicks were assigned randomly into five groups at 1 day of age, fifty chickens each. Group I was the control group, receiving no herbs; Group II was the positive herbal Group, birds in this group being given 0.5% powder of Astragalus root in feedstuff (0.5% ARP Group); Group III was the 0.5% SLASUP Group; Group IV was the 1% SLASUP group; and Group V was the 1.5% SLASUP Group. Chickens in the three SLASUP Groups were given SLASUP powder in the feedstuff. All birds in the five groups were immunised on day 7 and 21 with Newcastle disease (ND) vaccine. The blood samples on days 21, 28, 35, 42 and 49 were collected to determinethe ND virus (NDV) antibody titers, the contents of interleukin-2 (IL-2) and the immunoreactive fibronectin (IFN-γ) in the sera of 21, 35, 49day-old chicken. The results indicate that both 1 and 1.5% SLASUP can improve the immune functions of chickens significantly by increasing the antibody titers, IL-2 and IFN-γ contents. Stems and leaves in ultrafined powder of Astragalus may be clinically useful to boost the immune response in preventing infections such as ND in poultry farms.

BackgroundThe Toll-like receptor 4 (TLR4) pathway involves in the pathogen recognition and defense against infection in mammals. Considering that avian and mammalian TLR are differentially mediated, the action of a natural product on avian TLR4 pathway was unclear. High, medium and low doses of Astragalus polysaccharide (APS), were treated the chicken at 7-days-old age by gavage. The sIgA level in the intestinal fluid, the expression of chTLR4 mRNA/protein in bursa of Fabricius as well as the expression of downstream molecules of chTLR4 (chMyD88, chTRIF, chNF-κB, chIRF3, chIFN-β and chTNF-α) were measured on alternate days.ResultsThe content of sIgA and the chTLR4 mRNA expression/protein level were increased in non-dose-dependent manner after APS supplement. Also, the expressions of a subset of MyD88-independent pathway genes were more than MyD88-independent, in particular with low doses of APS supplement for 7 days.ConclusionsThese suggest that administration of APS activates chTLR4 pathway in bursa of Fabricius in MyD88-independent pathway. Meanwhile, low dose of APS shows better performance regarding the activation of chTLR4 and regulation of MyD88-independent pathway.

Effects of Selenylation Modification on Immune-Enhancing Activity of Garlic Polysaccharide

The garlic polysaccharide was modified by HNO3-Na2SeO3 method according to orthogonal design L9(34) to obtain nine selenizing garlic polysaccharides, sGPS1-sGPS9. Their effects on chicken peripheral lymphocytes proliferation in vitro were compared by MTT assay. The results showed that sGPSs could significantly promote lymphocytes proliferation, sGPS3, sGPS5 and sGPS6 presented stronger efficacy. In vivo experiment, 14-day-old chickens were injected respectively with sGPS3, sGPS5 and sGPS6 when they were vaccinated with ND vaccine taking unmodified GPS as control. The results showed that three sGPSs could significantly promote lymphocyte proliferation, enhance serum antibody titer, IFN-γ and IL-2 contents. These results indicated that selenylation modification could significantly enhance the immune-enhancing activity of GPS, sGPS6 possessed the best efficacy and could be as a candidate drug of immunoenhancer. Its optimal modification conditions were 400 mg of sodium selenite for 500 mg of GPS, reaction temperature of 70°C and reaction time of 6 h.
