In this study, one water-soluble polysaccharide, CPP, was purified from the root of Codonopsis pilosula. The immunomodulatory effect and the adjuvant potential of CPP on the cellular and humoral immune response of ICR mice against ovalbumin (OVA) were investigated. CPP was shown not to be lethal in vivo for mice in doses ranging from 0.5 to 4 mg. ICR Mice were immunized subcutaneously with 0.1 mg of OVA alone or with 0.1 mg of OVA dissolved in saline-containing aluminum hydroxide gel (Alum) (0.2 mg), QuilA (0.01 and 0.02 mg) or CPP (0.5, 1 or 2 mg) on days 1 and 15. Two weeks later (day 28), concanavalin A (ConA)-, lipopolysaccharide (LPS)-, and OVA-stimulated splenocyte proliferation, and OVA-specific serum antibodies were measured. CPP significantly enhanced the ConA-, LPS-, or OVA-induced splenocyte proliferation in the OVA-immunized mice especially at a dose of 1 mg (P<0.05 or P<0.01). The OVA-specific IgG, IgG1, and IgG2b antibody levels in serum were also significantly enhanced by CPP compared with OVA control group (P<0.05 or P<0.01). The results suggest that CPP could be a safe efficacious adjuvant for use in vaccines against both pathogens and cancer.

The purpose of this study was to evaluate the effects of dietary Soapwort Extract (SE) supplementation on laying performance, egg quality, some offal weights, some serum parameters, antioxidant status of liver and heart tissues and fatty acid composition of breast meat in chronic cold stressed Japanese quails. A total of sixty three 45-day-old Japanese quails were divided into three groups with three replicate. One group was fed with the corn-soybean based diet alone (control group) and the others were fed with the basal diet supplemented by 50 and 100 ppm SE. Cold stress was applied every night between 22.00 to 06.00 h as 7±1°C. SE supplementation had no significant effect on laying performance, offal weights and egg quality parameters with the exception of egg and albumen weights. Serum glucose, triglyceride, uric acid and aspartate aminotransferase (AST) levels were reduced in SE-50 group. While supplementation of 100 ppm SE decreased both liver and heart MDA levels, both amounts of SE increased the liver GSH-Px enzyme activity. Supplementation of 100 ppm SE decreased the levels of oleic acid and MUFA in breast meat, and it significantly increased the linoleic, linolenic and arachidonic acid, PUFA, total omega 6 and the ratio of omega 6/omega 3. It is displayed that SE supplementation to diets of laying quails exposed to cold stress can alleviate the detrimental effects of oxidative stress with no affecting on performance parameters. Toplam olarak 63 adet 45 günlük bıldırcın, 3 tekerrürlü 3 gruba ayrılmıştır. Gruplardan biri mısır-soya küspesine dayalı temel yem ile (kontrol grubu), diğer gruplar temel yeme 50 ve 100 ppm SE ilave edilen yemle beslenmişlerdir. Soğuk stresi her akşam 22.00-06.00 saatleri arasında kümes ısısı 7±1°C'ye düşürülerek uygulanmıştır. SE ilavesi performans, bazı organ ağırlıkları ile yumurta ve albümün ağırlığı hariç yumurta kalite parametrelerini etkilememiştir. Serum glikoz, trigliserit, ürik asit ve AST düzeyleri SE-50 grubunda düşmüştür. 100 ppm SE ilavesi karaciğer ve kalp dokularında MDA düzeylerini düşürürken, karaciğer GSH-Px enzim aktivitesi her iki SE grubunda artmıştır. 100 ppm SE ilavesi göğüs etinde oleik asit ve MUFA düzeylerini düşürürken, linoleik, linolenik ve araşidonik asit, PUFA, toplam omega 6 ile omega 6/omega 3 düzeylerini önemli ölçüde yükseltmiştir. RESUMO:Entre as diversas atividades biológicas relatadas para as saponinas, merecem destaque aquelas relacionadas ao aumento da resposta imune e a ruptura das membranas dos eritrócitos. No desenvolvimento de vacinas, ambas as propriedades exercem atividades antagônicas, contudo, as informações sobre as relações estrutura-atividade são relativamente escassas e, às vezes, conflitantes. O presente trabalho visa contribuir no estabelecimento das relações estruturais envolvidas com as atividades imunoadjuvante e hemolítica de saponinas triterpênicas. Para isso, foram selecionadas vinte saponinas de estrutura triterpênica, isoladas das espécies Aesculus hippocastanum, Dolichos lablab e Glycine max. A relação entre grupamentos substituintes do anel triterpênico e as atividades biológicas foi estudada mediante análise de agrupamentos e análise de componentes principais. Os resultados confirmam a importância da presença de açúcares em C-3 para a atividade hemolítica. Porém o efeito causado pela presença de uma hidroxila em C-16, de CH 2 OH em C-17, de uma acetila em C-22 e de um grupamento acila em C-21 sobre essa atividade parece ser mais acentuado. Já a presença de uma hidroxila em C-21, de uma metila em C-17 e de dois açúcares ligados à aglicona parece ser determinante para a atividade imunoadjuvante. Além disso, observa-se a existência de uma relação inversa entre as atividades hemolítica e imunoadjuvante.Unitermos: Saponinas, relação estrutura-atividade, atividade imunoadjuvante, atividade hemolítica, análise de agrupamentos, análise de componentes principais.ABSTRACT: "Study of the structure-activity relationship of adjuvant and/or hemolytic saponins by use of multivariate analysis". Among the various biological activities reported for saponins, those related with increase immune response and the destruction of the membrane of red blood cells deserve attention. In the development of vaccines, these properties exert antagonistic activities. However, the information about the structure-activity relationships is relatively scarce and sometimes contradictory. This paper aims to contribute to the establishment of structural relations involved with the hemolytic and adjuvant activities of triterpenic saponins by the analysis of the different groups linked to aglycone, with the use of multivariate statistical techniques. For this, we had twenty triterpenic saponins previously isolated from Aesculus hippocastanum, Dolichos lablab. and Glycine max species. Their adjuvant and hemolytic activities are available in the literature. These saponins were analyzed as groups linked to the triterpenic ring and biological activities resulting in an array of date that was submitted to cluster analysis and principal components analysis. The results confirm the importance of sugars in C-3 for hemolytic activity. However, the effect caused by the presence of a hydroxyl in C-16, CH 2 OH in C-17, acetyl in C-22 and acyl in C-21 on the hemolytic activity seems to be more important.
