Adhesion Mechanisms in Platelet Function

Abstract: Abstract-Platelet adhesion is an essential function in response to vascular injury and is generally viewed as the first step during which single platelets bind through specific membrane receptors to cellular and extracellular matrix constituents of the vessel wall and tissues. This response initiates thrombus formation that arrests hemorrhage and permits wound healing. Pathological conditions that cause vascular alterations and blood flow disturbances may turn this beneficial process into a disease mechanism t… Show more

“…Integrins also play an important role in cell signaling and most require activation before they are able to bind with high affinity to their ligands. Platelets express a number of b1 and b3 integrins, including a5b1 (VLA-5), a6b1 (VLA-6), a2b1 (GPIa/IIa, VLA-2, or CD49b/ CD29), and GPIIb/IIIa (aIIbb3) [12,13]. These molecules mediate platelet adhesion to ICAM and JAMs on leukocytes and endothelium and to extracellular matrix proteins, such as fibronectin, laminin, and collagen [12,13].…”

“…Instead, we considered the possibility that platelet-endothelial adhesion in this context is integrin mediated, as integrins have been reported to be critical in thrombus formation within injured vessels (36). We pretreated platelets with synthetic RGDS peptide, which competitively inhibits integrin binding to fibronectin and related proteins (37).…”

“…Whereas intermediate TXA 2 signaling supports platelet activation and aggregation specifically at low concentrations of thrombin, dense granule secretion and therefore aggregation in response to high concentrations of collagen markedly relies on TXA 2 synthesis and signaling [17].…”
