Adhesion dependent release of hepatocyte growth factor and interleukin-1 receptor antagonist from human blood granulocytes and monocytes: Evidence for the involvement of plasma IgG, complement C3 and ?2 integrin

Abstract: Plasma IgG, C3 derived complement activation fragments and leucocytes CD18 are involved in granulocyte adhesion to CA beads and hence the release of HGF and IL-1ra.

“…During Adacolumn treatment, the extracorporeal circulation results in complement fragments and immunoglobulins being deposited on the CA beads. These deposits induce the specific adsorption of granulocytes and monocytes, which express complement receptors (CD18/CD11b, Mac-1) and Fc receptors [19]. Since lymphocytes do not express these receptors, almost all CD3 1 lymphocytes pass through the CA bead column [20].…”

“…From the results of this study, we postulate that monocytes treated with CA beads act as immune suppressors, because intracellular TGF-b1, C5a, and hepatocyte growth factor (HGF) were either upregulated or released into the blood after treatment with CA beads (Fig. 4) [19,29]. C5a stimulation reduces the mRNA expression of IL-12 p35 and p40, IL-23 p19, and IL-27 p28 [28], which in turn induces the differentiation of naïve T cells into Th1 lymphocytes and cytotoxic Tlymphocytes, which subsequently activates NK cells.…”

“…Takeda et al [33] incubated human whole blood with the GMA carriers in in-vitro settings and measured significant amounts of IL-1 receptor antagonist (IL-1ra) and hepatocyte growth factor (HGF) released from granulocytes and monocytes that adhered to the carriers (Fig. 6); the amounts released were directly proportional to the number of cells that adhered to the carriers.…”

“…Therefore, the effects of granulocytapheresis on L-selectin and CXCR3 should minimize leukocyte accumulation in tissues. Fourth, more recently published data show that neutrophils and monocytes when adsorbed to the Adacolumn cellulose acetate carriers release large amounts of IL-1 receptor antagonist and hepatocyte growth factor that have strong anti-inflammatory and tissue repair actions [36].…”
