ADF/cofilin mediates actin cytoskeletal alterations in LLC-PK cells during ATP depletion

Abstract: Ischemic injury induces actin cytoskeleton disruption and aggregation, but mechanisms affecting these changes remain unclear. To determine the role of actin-depolymerizing factor (ADF)/ cofilin participation in ischemic-induced actin cytoskeletal breakdown, we utilized porcine kidney cultured cells, LLC-PKA4.8, and adenovirus containing wild-type (wt), constitutively active, and inactive Xenopus ADF/cofilin linked to green fluorescence protein [XAC(wt)-GFP] in an ATP depletion model. High adenoviral infectivit… Show more

“…These proteins are potent regulators of actin filament dynamics in eukaryotic cells and mediate the severing and depolymerization of actin. The actin-depolymerizing factor/cofilin family has been shown to play a crucial role in mediating microvillar breakdown and rapid cytoskeletal alterations in ischemic and ATP-depleted proximal tubule cells (26).…”

“…When ATP levels are not promptly restored, cofilin and ADP actin bind together and form filaments that associate into cylindrical aggregates, called rods. Rods have been observed in the nucleus and cell body of neurons on oxidative stress, 82 glutamate excitotoxicity, 83 and ischemia, 84 and they slowly contribute to neuronal dysfunction and death. Interestingly, the formation of actin rods implies not only an alteration of the synapses but it also contributes to an impairment of gene transcription.…”
