Adenovirus Precursor pVII Protein Stability Is Regulated By Its Propeptide Sequence

Abstract: Adenovirus encodes for the pVII protein, which interacts and modulates virus DNA structure in the infected cells. The pVII protein is synthesized as the precursor protein and undergoes proteolytic processing by viral proteinase Avp, leading to release of a propeptide sequence and accumulation of the mature VII protein. Here we elucidate the molecular functions of the propeptide sequence present in the precursor pVII protein. The results show that the propeptide is the destabilizing element targeting the precur… Show more

“…Both cell lines were grown in a humidified cell incubator at 37°C in the presence of 7% CO 2 . The stable HeLa cell line expressing the HAdV-5 pVII(Δ24)Flag protein was generated by integrating a codonoptimized pVII(Δ24)Flag cDNA (14) into the HeLa-Flp-In T-Rex cell line (generously provided by Stephen Taylor [44]). pVII(Δ24)Flag protein expression with doxycycline treatment and detection in whole-cell lysates were performed as reported previously (14).…”

“…Notably, MLTU encodes an abundant histone-like protein, pVII, which is proteolytically processed into the mature protein VII during the final steps of virion maturation (13). The mature protein VII [also known as pVII(Δ24) (14)] organizes the viral genome into a nucleosome-like structure (15)(16)(17), protects viral DNA from the cellular DNA damage response (18), and also can act as a protein regulating transcription (19,20).…”

“…The generated plasmids pcDNA3.1(Flag)PHPT1(wt) and pcDNA3.1(Flag)PHPT1(Spl) allow the expression of the PHPT1 proteins as N-terminal Flag-tagged fusion proteins in mammalian cells. Dominant negative Cullin1-5 (DN-Cul1-5) plasmids have been published before (Inturi et al, 2013).…”
