Adenovirus helper function for growth of adeno-associated virus: effect of temperature-sensitive mutations in adenovirus early gene region 2

Abstract: Adeno-associated virus (AAV) grows efficiently only in cells that are also infected with an adenovirus (Ad). We employed Ad mutants to determine which genes may be required for the AAV helper function. Two mutants of Ad type 5 (Ad5), Ad5ts125 and Ad5ts107, with temperature-sensitive lesions in the E72 DNA-binding protein coded by the Ad early region 2, were deficient for AAV helper functions at the nonpermissive temperature (40 degrees C). In contrast, Ad5ts149, with a temperature-sensitive lesion in the Ad ea… Show more

“…We identified BAAV in the same samples from Mexican cattle (Table 1) from which BAdV3 was recovered, supporting previous work that BAAV requires selected proteins derived from bovine adenovirus for its replication (Handa & Carter, 1979;Myers et al, 1980). A near-complete 4589 base sequence was assembled from 37 433 reads that shared 98 % nucleotide identity to the reference sequence (Gen-Bank AY388617).…”

“…Stocks of the Ad5 host-range mutants hr6 and hrl3 (provided by J. Williams), and sub316 (provided by T. Shenk) were grown in 293 cells. Stocks of the temperature-sensitive mutants AdSts215 (provided by W. Doerfler) and AdStsl49 (provided by H. Ginsberg) were prepared in KB cells as described previously (39). Herpes simplex virus types 1 and 2 (HSV-1 and HSV-2) were provided by I. Hay, and new stocks were grown on Vero cells.…”

“…In addition, Ostrove and Berns [66] reported that E1B is required for rescue of AAV DNA from the integrated state in human Detroit 6 cells but not for DNA replication. For the latter process, early region 4 appears to be necessary [67,68], and region E2A and VAI RNA are said to be involved in post-transcriptional regulation of capsid protein synthesis [69]. The requirement for VAI RNA during replication of AAV does not seem to be absolute.…”
