Adenosine 5'‐monophosphate‐activated protein kinase‐dependent mTOR pathway is involved in flavokawain B‐induced autophagy in thyroid cancer cells

Abstract: Flavokawain B (FKB), a natural kava chalcone, shows potent antitumor activity in various types of cancer, although the mechanism of action remains unclear. In this study, we report that FKB has profound effects on the metabolic state of human thyroid cancer (TCa) cells, leading to high autophagy flux through upregulation of AMP‐activated protein kinase, which in turn inhibits mTOR and activates Beclin‐1 in TCa cells. We further report that the autophagy induced by FKB plays a prosurvival role in TCa cells both… Show more

“…During cytotoxicity of FKB, FKB induced autophagy by increasing LC3B-II and Beclin 1 expression and decreasing p62 expression. 121 In this study, Beclin 1 was activated by activating AMPK in a similar manner as in other studies. The antitumor effect of FKB was improved by inhibition of autophagy through 3-MA or cotreatment with Beclin 1 siRNA and ATG5 siRNA.…”

“…GFP-MAP 1LC3B (pBABEpuro, 22405)-expressing vectors were purchased from Addgene (Cambridge, MA, USA). The constructs, transfections, and lentiviral infections of GFP-MAP 1LC3B that we used have been described previously [24]. After treatments, we calculated the number of autophagic cells expressed as GFP-MAP 1LC3B dots (positive cells as ≥ 3 dots).…”

“…Flavokavain B was reported to have significant anti-tumor effects on several carcinoma cell lines both in vitro and in lab animal models. Similar to flavokavain A, flavokavain B was reported to inhibit cancer cell proliferation [ 118 ], metastasis [ 128 , 129 ] and angiogenesis [ 130 ], and induce apoptosis [ 128 , 129 , 131 , 132 , 133 , 134 , 135 , 136 ], G2/M [ 128 , 132 , 136 , 137 ], G0/G1 [ 138 ] cell cycle arrest, autophagy [ 134 , 139 , 140 , 141 , 142 ], and the immune responses [ 143 ]. Unlike flavokavain A, flavokavain B induced G2/M cell cycle arrest in cancer cells regardless of the p53 status [ 128 , 131 , 132 , 134 , 136 , 137 ], which is intriguing given their similar structures ( Figure 2 ).…”
