Adeno-associated Virus Vector-mediated Interleukin-10 Induction Prevents Vascular Inflammation in a Murine Model of Kawasaki Disease

Abstract: Kawasaki disease (KD), which is the leading cause of pediatric heart disease, is characterized by coronary vasculitis and subsequent aneurysm formation. Although intravenous immunoglobulin therapy is effective for reducing aneurysm formation, a certain number of patients are resistant to this therapy. Because interleukin-10 (IL-10) was identified as a negative regulator of cardiac inflammation in a murine model of KD induced by Candida albicans water-soluble fraction (CAWS), we investigated the effect of IL-10… Show more

“…Echocardiography in CAWS-injected mice indicates a marked decrease of cardiac function, which can be restored by IL-10 supplementation 193 . IL-10 is a potent anti-inflammatory cytokine that might improve the outcome of CAWS-induced vasculitis by inhibiting the release of pro-inflammatory mediators, such as TNF and IL-1β, from tissue-infiltrating innate immune cells 194 .…”

“…Real-time RT-PCR analysis was performed as described previously 17,47,48 . Total RNA was prepared from the parietal and visceral peritoneum using ISOGEN (Nippon Gene Co., Ltd., Toyama, Japan) according to the manufacturer’s instructions.…”

“…Western blot analysis was performed as described previously 17,47,48 . Briefly, cell lysates were prepared using RIPA buffer (20 mM Tris, 2.5 mM EDTA, 1% Triton X, 10% glycerol, 1% deoxycholic acid, 0.1% SDS, 50 mM NaF, and 10 mM Na 4 P 2 O 7 ·10H 2 O), and subjected to SDS–PAGE.…”

“…IL-10 has long been shown to be elevated during acute KD (58,59). This is likely a natural anti-inflammatory response, as recent studies show supplementing IL-10 via an AAV vector protected against aneurysms in the Candida albicans murine model (60). IL-10 is produced by myeloid dendritic cells and regulatory B cells, and recently has been shown to drive plasmablast responses (discussed later) (61).…”
