Adaptation of the cerebrocortical circulation to carotid artery occlusion involves blood flow redistribution between cortical regions and is independent of eNOS

Abstract: Authors' contributions: ZB, PS and BH planned and supervised the project; BH and AI 13 introduced laser-speckle imaging for studying cerebrocortical microcirculation in mice and 14 optimized the experimental procedures in preliminary experiments; AP, LH, AI and DS 15 performed the experiments; AP, LH, DS and ÉR evaluated the experiments and analyzed the 16 data; AP and ÉR prepared the figures and tables; AP, LH, PS and ZB wrote the manuscript.

“…Following the cannulation of the left femoral artery, the trachea was exposed in order to improve the breathing of the mice through an intratracheal cannula. Subsequently, the carotid sheath was gently dissected with particular care to preserve the vagus nerve [17]. For the later induction of CAO, a ligature with a loose knot was placed around the left common carotid artery.…”

“…We waited until atipamezole exerted its effect (ca. 10 min) [17], then 10 min were allowed to acquire baseline data of CoBF and blood pressure. Thereafter, the left common carotid artery was occluded by tightening the loose knot around the vessel [17].…”

“…The probe implantation involves permanent ligation of an artery, most commonly the left carotid in mice, which is a significant perturbation to the circulation 6. The weight of the probe can compromise animal welfare,7 as well as the requirement for single housing for the purposes of data collection 8, 9.…”

“…The probe implantation involves permanent ligation of an artery, most commonly the left carotid in mice, which is a significant perturbation to the circulation. 6 The weight of the probe can compromise animal welfare, 7 as well as the requirement for single housing for the purposes of data collection. 8,9 Although telemetry systems compatible with group housing are becoming available for mice, a relatively high cost and requirement for major surgery remain important limiting factors.…”
