ADAMTS19 Suppresses Cell Migration and Invasion by Targeting S100A16 via the NF-κB Pathway in Human Gastric Cancer

Abstract: A Disintegrin and Metalloproteinase with Thrombospondin motifs 19 (ADAMTS19) has been reported to participate in the pathogenesis of solid cancers. However, its role in gastric cancer (GC) remains undocumented. Using immunohistochemistry (IHC) staining and quantitative real-time polymerase chain reaction (qRT-PCR) on GC tissues and adjacent normal tissues, we found that ADAMTS19 was downregulated in GC tissues (IHC: p < 0.001; qRT-PCR: p = 0.017). Further investigation revealed that ADAMTS19 correlated with… Show more

“…The molecular mechanisms of S100A16 involving in the tumor metastasis are diverse in various malignant tumors, including pancreatic cancer ( Fang et al, 2021 ; Zhuang et al, 2021 ), leukemia ( Zhang et al, 2019 ), breast cancer ( Zhou et al, 2014 ), and so on. In gastric cancer, decreased S100A16 expression is involved in miR-6884-5p and ADAMTS19 (A Disintegrin and Metalloproteinase with Thrombospondin motifs 19) inhibition of GC cells migration, invasion, and EMT ( Lv et al, 2020 ; Jiang et al, 2021 ), however, the role of S100A16 itself in GC metastasis has not been fully clarified yet.…”

“…Among these genes, we identified S100A16 using multiple criteria, including its association with glioma, functional validation, and overall survival analysis. S100A16 is involved in various tumors, such as colorectal cancer, bladder cancer, pancreatic cancer, lung adenocarcinoma, cervical cancer, leukemia, and gastric cancer [37][38][39][40][41][42][43]. According to the CGGA database, S100A16 closely correlates with the survival rate of patients with primary gliomas.…”

“…Immunohistochemistry and immuno uorescence assay. The biotin-streptavidin horseradish peroxidase (HRP) detection system (ZSGB Bio, China) was used for IHC staining as previously described 22 . In brief, primary rabbit antibody against ADAMTS12 (24934-1-AP, Proteintech, China, 1:1200), VEGF (19003-1-AP, Proteintech, China, 1:2000), cleaved caspase-3 (Cell Signaling Technology, Massachusetts, USA, 1:800), Ki67 (Cell Signaling Technology, Massachusetts, USA, 1:300), and CD31 (Cell Signaling Technology, Massachusetts, USA, 1:200) were incubated with the specimens at 4°C overnight.…”

“…For immuno uorescence staining, para n-embedded sections were de-para nized by xylene and retrieved via EDTA (1 mM EDTA, 0.05% Tween 20, pH 8.0) for 20 minutes at boiling temperature. The follow-up procedure was the same as in the previous study 22 .…”
