ADAM17 promotes the invasion of hepatocellular carcinoma via upregulation MMP21

Abstract: Background The upregulation of ADAM17 has been reported to be associated with invasion and metastasis in various tumors, however the molecular mechanism of ADAM17 in the progression of hepatocellular carcinoma (HCC) remain to be clarified. Human matrix metalloproteinase 21 (MMP21), the newest member of the MMP gene family, has been suggested to play an important role in embryogenesis and tumor progression. So far, nothing is known about the relationship between ADAM17 and MMP21. Methods In this study, the ex… Show more

“… 24 The matrix metalloproteinases (MMPs) also participate in invasiveness of HCC cells, so that ADAM17 induces MMP-21 expression that is in favor of increasing HCC progression. 25 As mentioned, lncRNAs regulate HCC progression. Notably, microRNAs (miRNAs) also affect malignancy of HCC cells.…”

“…Although we did not measure these other proteins directly, we hypothesize that their rate of cleavage might also be increased in an ERK‐dependent manner in LCH cells. Overactive TACE activity has been observed and reported to contribute to various inflammatory disorders and cancers 64,65 . Not surprisingly, there have been efforts to target TACE as a therapeutic, but safety and toxicity concerns have prevented its clinical progression 66 …”

“…Moreover, ADAM17 has been described to be involved in EGF-R- mediated IL-6 synthesis and tumorigenesis in colorectal cancer [ 37 ]. In hepatocellular carcinoma (HCC), ADAM17 seems to be involved in the development of HCC invasion and metastasis [ 38 ]. However, as the regulation and pathophysiology of ADAM17 in cancer and especially in tumor-educated platelets is complex, further elucidation is warranted.…”
