ADAM10 and ADAM17 as Biomarkers Linked to Inflammation, Metabolic Disorders and Colorectal Cancer

Abstract: ADAM10 and ADAM17 have a role in inflammation and diseases associated with inflammation, such as diabetes, cardiovascular diseases (CVD) or cancer, e.g., colorectal cancer (CRC). The aim of this study was to evaluate whether ADAM10 and ADAM17 could be biomarkers of CRC. To achieve this goal, CRC tumors and a surgical margin from 72 patients with CRC were collected. The concentration of ADAM proteins was measured by the ELISA method. Results were analyzed statistically and compared with selected clinical parame… Show more

“…These observations were confirmed by recent analyses showing that serum levels of ADAM10 and ADAM17 are higher in patients with CRC, and that this level correlates with the degree of differentiation of tumor cells and the presence of distant metastases [ 55 ]. The association of ADAM17 with CRC arising on the basis of inflammatory changes may also be confirmed by one recent study, where a higher concentration of ADAM17 was observed in the CRC tissue of patients with concomitant diabetes and cardiovascular diseases [ 56 ]. In addition, ADAM12 and ADAM28 proteins are involved in the pathogenesis of CRC, promoting tumorigenesis by activating pathways related to IGF.…”

“…To assess the effects of adipose tissue-specific ADAM10 deletion on adipokine release, we measured serum Adiponectin and Leptin serum levels and Adiponectin/Leptin ratio, the latter an index of adipose tissue inflammation [ 13 ].…”

“…Previous studies have demonstrated that ADAM sheddase family, through their activation of the TNF-a signalling, are involved in inflammation, and indirectly to metabolic disorders, insulin resistance, and development of malignancy [ 13 , 18 , 19 ].…”

“…Julia and colleagues suggested that TNFR1-dependent tumor cell-induced endothelial cell death, tumor cell extravasation, TNFinduced necroptosis, and subsequent metastatic seeding rely on the activation of the membrane-bound metalloprotease ADAM17 41 . In addition, all of these targets serve as prognostic biomarkers for many other types of cancers, such as ovarian cancer, colorectal cancer, hepatocellular carcinoma, and kidney cancer [42][43][44][45] . However, the biological function of these genes in CCA is still unclear, and further investigation is needed to explore the potential mechanisms of these target genes in the future.…”
