Acute Toxicity Assessment: Macroscopic and Ultrastructural Effects in Mice Treated with Oral Tetrodotoxin

Abstract: Tetrodotoxin (TTX) is an extremely toxic marine compound produced by different genera of bacteria that can reach humans through ingestion mainly of pufferfish but also of other contaminated fish species, marine gastropods or bivalves. TTX blocks voltage-gated sodium channels inhibiting neurotransmission, which in severe cases triggers cardiorespiratory failure. Although TTX has been responsible for many human intoxications limited toxicological data are available. The recent expansion of TTX from Asian to Euro… Show more

“…However, animals treated orally with a single TTX dose from 125 to 500 µg/kg b.w. showed liquid and gas accumulation in the stomach while ultrastructural changes were observed in the spleen, liver and intestine two hours after oral administration of TTX to mice at doses of 500 µg/kg [22]. Recently, the first report on the chronic toxicity of TTX after exposure of mice to low oral doses of toxin following the internationally adopted guidelines of the Organisation for Economic Co-operation and Development (OECD) has been released [23].…”

“…Stomach samples preparation for transmission electron microscopy was performed according to previously described procedures [ 35 , 57 , 58 ]. Concisely, organ samples (1 mm 3 ) were fixed by immersion (2.5% glutaraldehyde in phosphate buffered saline (PBS) containing in mM: 137 NaCl, 8.2 Na 2 HPO 4 , 1.5 KH 2 PO 4 and 3.2 KCl) for 2 h at 4 °C and rinsed three times with PBS.…”

“…However, EFSA also highlighted the need to re-evaluate the seafood safety risk to consumers since recently reported results pointed to potential harmful effects of chronic low oral doses of TTX. This requires further and detailed studies, especially considering the possible synergies between different marine biotoxins with the same mechanism of action [ 43 , 45 , 56 ].…”
