Acute Phase Protein α1-Acid Glycoprotein Interacts with Plasminogen Activator Inhibitor Type 1 and Stabilizes Its Inhibitory Activity

Abstract: ␣ 1 -Acid glycoprotein, one of the major acute phase proteins, was found to interact with plasminogen activator inhibitor type 1 (PAI-1) and to stabilize its inhibitory activity toward plasminogen activators. This conclusion is based on the following observations: (a) ␣ 1 -acid glycoprotein was identified to bind PAI-1 by a yeast two-hybrid system. Three of 10 positive clones identified by this method to interact with PAI-1 contained almost the entire sequence of ␣ 1 -acid glycoprotein; (b) this protein formed… Show more

“…59 One could speculate that a-1-AGP delays thrombin generation by playing the same role with other coagulation inhibitors belonging to the serpin family. Conversely, a-1-AGP has been observed to shorten aPTT 60 and to contribute to the cellular initiation of coagulation by inducing monocyte expression of TF.…”

“…There was also a significant induction of ␣1-acid glycoprotein mRNA, which we recently described to form a complex with active PAI-1 and stabilize its activity. 23 The stimulating effect of exosomes on PAI-1 synthesis was further evidenced by measuring the amount of PAI-1 released to culture media from these cells, as determined by enzyme-linked immunosorbent assay (Figure 2A). To determine the molecular level at which PAI-1 expression can be regulated, we used a transient transfection approach to ask if exosome components can influence the transcription of the PAI-1 promoter.…”

“…(Boncela et al, 2001); however, in that study AGP did not interfere with PAI-1 activity. As previously mentioned, serum AGP concentrations increase during late gestation in the pig and are highest at birth (Lampreave and Piñeiro, 1992), then rapidly decline postnatally Heegaard et al, 2013).…”
