Acute Inflammation Following Intraperitoneal Injection of Antigen into Actively Sensitised Rats

Abstract: Rats actively sensitised to bovine serum albumin (BSA) in Freund’s complete adjuvant were challenged by intraperitoneal injection of BSA. By washing out the peritoneal cavities at different times it was possible to divide the changes that occurred into two phases. The first was characterised by the release of histamine and slow reacting substance of anaphylaxis (SRS-A) with an associated extravasation of plasma proteins. The second phase was characterised by an increase in neutrophilic polymorphonuclear cells … Show more

“…The early phase of the reaction, perhaps mediat ed by IgE or IgG»., [2,10], involves the re lease of histamine and SRS-A with an asso ciated extravasation of plasma proteins. The later phase of the reaction, perhaps me diated by humoral antibody and comple ment, consists of an infiltration of inflam matory cells, especially of neutrophils [12].…”

“…The methods were essentially as described pre viously [12]. Briefly, rats were sensitised by inject ing 50 /ig BSA in Freund's complete adjuvant (FCA) intradermally, followed 28 days later by 100 ug BSA in saline subcutaneously.…”

“…The former is believed to be established by vasoactive/bronchoactive mediators with a subsequent increase in the vas cular permeability and airway resistance [2], The latter is characterized by an intense cellular infiltration, mainly eo sinophils [3]. Since the phenomenon of chemotaxis is one of the proposed mechanisms by which eosinophils may ac cumulate in a tissue [4], some endogenous eosinophil chemotaxins such as eosinophil chemotactic factor of anaphy laxis [5] and mediators such as C5a [6], leukotriene B4 (LTB4) [7] -preferentially PAF-acether [8] -are potential contributors to tissue eosinophil infiltration.…”
