Activity of clindamycin with primaquine against Pneumocystis carinii in vitro and in vivo

Abstract: The combination of primaquine with clindamycin is effective in both in vitro and in vivo models of Pneumocystis infection. Primaquine alone at concentrations from 10 to 300 ,ug/ml reduced the numbers of organisms in cultures to less than 7% of control. Significant inhibition was observed down to 0.1 ,ug/ml. Clindamycin at 5 ,ug/ml was ineffective alone. Combinations of clindamycin and primaquine in culture at various concentrations were effective, but there was no evidence of true synergy. In rats with establi… Show more

“…This drug is already in clinical assays for the oral treatment of visceral leishmaniasis [41], with phase II trials in India and Kenya being quite encouraging [229,230]. Physicochemical analysis of the interaction of sitamaquine with phospholipid monolayers and sterols at the air-water interface showed interactions only to occur in the presence of anionic phospholipids.…”

“…The combination of clindamycin and primaquine was shown to be effective in animals (145) and has been used successfully for the treatment of Pneumocystis pneumonia (98,160,180). Other 8-aminoquinolines have been evaluated for treatment and prophylaxis in animal models and appear to be very effective (12a, 12b …”
