Activity Guided Fractionation of Arum italicum Miller Tubers and the LC/MS-MS Profiles

Abstract: Abstract:In this study, the polar extract of Arum italicum Miller tubers was fractionated by 'biological activity guided fractionation' method. MCF-7 (human breast adenocarcinoma) and A549 (non-small lung cancer) cancer cell lines were used to screen cytotoxic activity. Possible antiproliferative and apoptotic effects of each subfraction were evaluated by in vitro. As a result, E2 were cytotoxic against A549 cell lines while E3 and E6 showed good cytotoxic activities against both cancer cell lines. These three… Show more

“…The results obtained in both RP-HPLC-UV and LC-MS /MS are similar to the results of study by Ağalar et al (2017). Ağalar et al (2017) stated that ferulic acid and caffeic acid were present in the leaves of plants belonging to the Araceae family, also p-coumaric acid in the seeds of some species of the same family. Hydroxycinnamic acids containing ferulic acid, caffeic acid, and p-coumaric acid prevent metastasis or invasion of cancer cells (Ağalar et al, 2017).…”

“…While ferulic acid, p-coumaric acid, epicatechin, vanillin, caffeic acid, phydroxy benzoic acid and salicylic acid are identified as Phenolic components defined and measured in both RP-HPLC-UV and LC-MS/MS are compatible with each other except for minor differences. Ağalar et al (2017) reported that the whole tuber part of Arum italicum has hydroxycinnamic acid derivatives such as ferulic, caffeic and p-coumaric acid in analysis with LC-MS/MS. The results obtained in both RP-HPLC-UV and LC-MS /MS are similar to the results of study by Ağalar et al (2017).…”

“…Depending upon the fact that no pertinent ion derived due to complete loss of hexosyl unit (−162 amu) was detected, suggested that sugar was C-linked. Thus (4d) was tentatively named as apigenin-C-hexoside-C-hexoside 56,59 . (Fig.…”

“…[51,52] Compound (7) at R t , 1.3 min yielded a deprotonated ion [M-H] − at 387 and further MS-fragment ions at 341, 323 and 233 were also identified in spectrum. A characteristic fragment at 179 revealed that the compound contains caffeic acid, [53] while the fragments at 161 and 131 indicated the cleavage of H 2 O and CH 2 O from the fragment at 179. However, compound (7) was identified tentatively as caffeic acid derivative (Figure 2 and Table 4).…”
