Active targeting of orthotopic glioma using biomimetic liposomes co-loaded elemene and cabazitaxel modified by transferritin

Abstract: Background Effective treatment of glioma requires a nanocarrier that can cross the blood–brain barrier (BBB) to target the tumor lesion. In the current study, elemene (ELE) and cabazitaxel (CTX) liposomes were prepared by conjugating liposomes with transferrin (Tf) and embedding the cell membrane proteins of RG2 glioma cells into liposomes (active-targeting biomimetic liposomes, Tf-ELE/CTX@BLIP), which exhibited effective BBB infiltration to target glioma. “…The liposomes are promising transporters to deliver antitumoral drugs to GBM [ 27 , 43 , 44 , 45 , 46 , 47 ]. However, there is evidence that lymphatic drainage of the brain is impaired in animals with GBM [ 57 , 58 ].…”

“…Liposomes are promising transporters to deliver anti-cancer drugs to GBM [ 27 , 43 , 44 , 45 , 46 , 47 ]. They offer numerous advantages, such as the similarity of the liposomal bilayer with the cell membrane; liposomes provide low drug toxicity, stability, and increased circulation time as well as targeting liposomes by modifying with antibodies, and polypeptides [ 43 , 48 , 49 ].…”

“…Therefore, the development of effective methods bypassing the IBBB at the border of GBM is the first step in the GBM therapy. Liposomes are promising nanocarriers to deliver anti-cancer drugs to GBM [ 37 , 68 , 69 , 70 , 71 ]. Therefore, in the final step, we studied the 1268 nm laser-induced BBBO for the delivery of liposomes into GBM.…”

“…In glioma therapy, TMZ and siEGFR encapsulated within NPs must be released into the glioma cells in order to exhibit a synergistic anticancer effect. Researchers have modified a targeting ligand-transferrin (Tf) liposome to specifically recognize transferrin receptors (TfR), increasing brain drug delivery ( Li et al, 2021 ). In this study, Tf was modified on the surface of PEG-PAE(SS)/TMZ@siEGFR to increase the BBB penetrating efficiency and the glioma distribution of drugs.…”
