Active Principle of Swine Prostate Extract:I. Isolation of Active Principle Activating Prostatic Acid Phosphatase and Its Effect on Testosterone Uptake of the Prostate in Castrated Rats.

Abstract: ABSTRACT-There have been several reports concerning the therapeutic effect of an extract from animal prostates on benign prostatic hypertrophy. Previously, we re ported that the swine prostate extract (PE) had the activity to enhance human prosta tic acid phosphatase (PAPase) activity in vitro, and to increase the muscular tonicity of the urinary bladder by directly acting upon vesical muscles, suggesting that PE have an activity to elevate the intravesical voiding pressure in vivo. In the present study, it wa… Show more

“…L-tartaric acid and ammonium molybdate are known as high molecular weight (>45 kDa) acid phosphatase inhibitors (van Etten 1982). But low molecular weight acid phosphatases are also sensitive on the addition of above mentioned inhibitors (Yoshida et al 1991, Granjeiro et al 1997. In our study L-tartaric acid and ammonium molybdate had also inhibitory effect on AcP activity from Russian sturgeon spermatozoa.…”

“…We the prevention of the recovery in the tissue weight. This effect of PPE was probably due to its ability to suppress the excessive testosterone-uptake into the prostate as evi denced in the preceding paper (7). PPE is a peptide 7, and the agent should display anti androgenic activity through a mechanism differ ent from that of a steroid hormone such as gestagen.…”

“…PPE activated PAPase in a dose-dependent fashion as well as recovered PAPase activity from the inhibition by L-tartaric acid. Fur thermore, the oral administration of PPE sup pressed uptake of 3H-testosterone into the prostate of castrated rats (7). These results suggested that PPE was one of the active prin ciples of PE for the therapeutic effect on prostatic hypertrophy.…”
