Active immunization of hamsters against experimental infection with Borrelia burgdorferi

Abstract: The immunogenicity of a single dose of a whole-cell vaccine of inactivated Borrelia burgdorferi1HSF (human spinal fluid isolate) was evaluated. Syrian hamsters were vaccinated subcutaneously and challenged by the intraperitoneal injection of 1,000 50% infectious doses of B. burgdorferi HSF 30 or 90 days postvaccination.Animals were sacrificed 14 days after challenge, and the kidneys and spleens were examined for spirochetes by cultural procedures. At 30 days postvaccination, 86 to 100% protection against infec… Show more

“…B. burgdorferi 297 was obtained from Russell C. Johnson (University of Minnesota, Minneapolis). The strain was originally isolated from human spinal fluid (24) and has been maintained by passage in Barbour-Stoenner-Kelly medium (BSK) and hamsters (10)(11)(12)(13)(14)19).…”

“…B. burgdorferi 297 was obtained from Russell C. Johnson (University of Minnesota, Minneapolis). The strain was originally isolated from human spinal fluid (26) and has been maintained by passage in modified Barbour-Stoenner-Kelly medium (BSK; 5) and hamsters (11)(12)(13)(14)(15)21). The hamster-passed spirochetes were grown in BSK at 35°C for 5 days.…”

“…Acquired immunity in the vertebrate host plays an important role in the epidemiology of Lyme disease (Johnson et al ., 1986a,b; Kurtenbach et al ., ; Piesman et al ., ; Liang et al ., ). One Borrelia antigen that is particularly important for the pathogen's interaction with the vertebrate immune system is outer surface protein C (OspC) (Radolf and Caimano, ).…”
