Active DNA Topoisomerase IIα Is a Component of the Salt-stable Centrosome Core

Abstract: Recently, we reported that the monoclonal antibody specific for human DNA topoisomerase II␣, Ki-S1, stains not only the nuclei of human A431 cells but also extranuclear structures suggestive of centrosomes (Meyer, K. N., Kjeldsen, E., Straub, T., Knudsen, B. K., Kikuchi, A., Hickson, I. D., Kreipe, H., and Boege, F. (1997) J. Cell Biol. 136, 775-788). Here, we confirm colocalization of Ki-S1 with the centrosomal marker ␥-tubulin. In addition, we show labeling of centrosomes by peptide antibodies against the N … Show more

“…TopoII signal was also observed on chromosomes, consistent with previous studies (20,43). It should be noted that a strong TopoII signal was present at the spindle poles, consistent with previous studies describing the presence of this protein in centrosomes (6). Whereas both TopoII and hCAP-E remained present on chromosomes in hCAP-D2-depleted cells, the chromosomal localization of hCAP-H was strongly diminished (Fig.…”

“…Nonconfluent cells (ϳ10 6 ) were treated with nocodazole (5 g in 10 ml of culture medium) for 6 h. The cells were recovered by mitotic shake off and sedimented by centrifugation (450 ϫ g for 3 min). The cell pellet was resuspended in 2 ml of culture medium and mixed with 3 ml of tap water.…”

“…Although Topo II is a major component of mitotic chromosomes and is generally considered to be a chromatin protein, one recent study has shown that Topo IIα can be detected as a salt-stable component of centrosomes in a number of human cell lines (Barthelmes et al, 2000). Although that study did not propose a definitive function for Topo II at the centrosome, our results suggest that this centrosomal Topo II could have a role in enabling the arms of metaphase chromosomes to detach from centrosomes and assume their classical condensed structure at the metaphase plate.…”

“…DNA topoisomerase II, referred to as TOP2, is essential for cell survival and plays a role in replication, transcription, chromosome condensation and segregation [18,19,20,21,22]. It allows one double-stranded DNA segment to pass through another, thus altering DNA topology.…”
