Activation of uterine intraepithelial γδ T cell receptor‐positive lymphocytes during pregnancy

Abstract: Intraepithelial lymphocytes (IEL) of the uterus of non-pregnant sheep were analyzed by single- and two-color flow cytometry. Very few lymphocytes carrying classical B and T cell markers (CD5, surface immunoglobulin) were detected in the uterine epithelial cell suspensions and all IEL expressed the CD8 surface marker although with varying intensities. Three distinct subpopulations were identified including a major (46-56%) population of CD8+CD45R- gamma delta T cell receptor (TcR)-negative cells and approximate… Show more

“…15,[51][52][53][54] Furthermore, there is evidence suggesting that the early pregnancy human decidua is actually a site of extrathymic maturation of Vd1 T cells. 55,56 This enrichment of cd T cells at the maternal/fetal interface and their mysterious role in pregnancy is not unique to humans as the same has been observed in sheep 57 and mice 58 , suggesting a central conserved role of cd T cells involved in pregnancy-related immune changes. In women with healthy pregnancies, the changes are not only found at the maternal/fetal interface since the number of Vd1 T cells in circulation have also been reported to be increased in relation to the Vd2 subset.…”

“…Although there was an increase in lymphocyte concentrations in the involution secretion with advancement of involution, most of the lymphocytes resided in the glandular tissues and most of the intraepithelial lymphocytes were CD8 + . Since there were very few γδTCR + cells in the glandular epithelium, it is possible that most of the CD8 + intraepithelial lymphocytes belong to the CD8 + CD45R + γδTCR V subpopulation reported by Meeusen et al (1993). The phagocytic leucocytes, both neutrophils and macrophages, were destined for the alveolar and ductal lumina.…”
