Activation of Trk Neurotrophin Receptor Signaling by Pituitary Adenylate Cyclase-activating Polypeptides

Abstract: Pituitary adenylate cyclase-activating polypeptide (PACAP), a neuropeptide that acts through G proteincoupled receptors, exerts neuroprotective effects upon many neuronal populations. However, the intracellular signaling mechanisms that account for PACAP's trophic effects are not well characterized. Here we have tested the possibility that PACAP uses neurotrophin signaling pathways. We have found that PACAP treatment resulted in an increase in TrkA tyrosine kinase activity in PC12 cells and TrkB activity in hi… Show more

“…Many receptor tyrosine kinases (RTKs) undergo transactivation by agonists of GPCRs (Daub et al, 1997;Lee and Chao, 2001;Oak et al, 2001;Lee et al, 2002b;Fischer et al, 2004;Zahradka et al, 2004) and our observations that antagonism of adenosine A2a receptors in spinal cord cultures leads to a reduction in TrkB activation complement these results. They imply that under physiological conditions, adenosine A2a and TrkB receptors endogenously interact and active adenosine A2a receptors contribute to the basal level of TrkB activation.…”

“…Previous studies have highlighted a role for Src family kinases in TrkA and epidermal growth factor receptor transactivation (27)(28)(29). Here, AEA-activated Src family kinases, which was reversed by the selective Src family kinase inhibitor PP2 (Fig.…”

“…Trk activation is associated with activation of a variety of G-protein coupled receptors (GPCRs) including A2a adenosine receptors (Lee and Chao, 2001), PAC1 receptors for the neuropeptide pituitary adenylate cyclase activating peptide (PACAP) (Lee et al, 2002), and endocannabinoid receptors (Berghuis et al, 2005). GPCRdependent Trk transactivation differs from neurotrophin-dependent Trk activation in one important respect.…”
