Activation of the Potato Tuber ADP-glucose Pyrophosphorylase by Thioredoxin

Abstract: The potato tuber (Solanum tuberosum L.) ADP-glucose pyrophosphorylase (ADP-GlcPPase) catalyzes the first committed step in starch biosynthesis. The main type of regulation of this enzyme is allosteric, and its activity is controlled by the ratio of activator, 3-phosphoglycerate to inhibitor, P i . It was reported (Fu, Y., Ballicora, M. A., Leykam, J. F., and Preiss, J. (1998) J. Biol. Chem. 273, 25045-25052) that the enzyme was activated by reduction of the Cys 12 disulfide linkage present in the catalytic sub… Show more

“…We were able to detect both subunits of ADP-glucose pyrophosphorylase, an enzyme known to be regulated by Trx in amyloplasts as well as chloroplasts, (18,48). Taking into consideration the previously unrecognized targets of starch metabolism listed in Table 2, it appears that Trx controls this process at several levels in plastids, including transport, breakdown, and synthesis.…”

“…First, enzymes shown to be regulated by Trx reside in the organelle, namely, ADP-glucose pyrophosphorylase (18), NADP-glucose 6-phosphate dehydrogenase (19,20), and ␣-glucan, water dikinase (21). Second, analyses of the constituent proteins (proteome) of amyloplasts have revealed the presence of a number of other enzymes that, in chloroplasts, are linked to Trx (refs.…”

“…Moreover, the enzyme responsible for the first committed step in starch biosynthesis, ADP-glucose pyrophosphorylase (EC 2.7.7.27), has recently been shown to be redox-regulated (37)(38)(39). When searching the databases, we found the regulatory site sequence of potato GWD conserved in other GWD homologs (Fig.…”

“…On the contrary, ADP-Glc PPases from wheat, maize, and rice endosperm, as well as from potato tuber and cassava root, are regulated by 3-PGA and P i (3,(11)(12)(13)(14). In addition to the regulation by 3-PGA and P i , a redox control has also been proposed for potato tuber ADP-Glc PPase (15)(16)(17).…”
