Activation of the Notch1/STAT3/Twist signaling axis promotes gastric cancer progression

Abstract: Gastric carcinoma is one of the most common malignancies and a lethal cancer in the world. Notch signaling and transcription factors STAT3 (signal transducer and activator of transcription 3) and Twist regulate tumor development and are critical regulators of gastric cancer progression. Herein, the relationship among Notch, STAT3 and Twist pathways in the control of gastric cancer progression was studied. We found that Twist and phosphorylated STAT3 levels were promoted by the activated Notch1 receptor in huma… Show more

“…The mortality rate of patients with GC is increasing in developing countries, particularly in East Asian countries (30). The overall 5-year survival rate remains low; ranging between 20-30% for patients past the early stage of the disease at the time of their initial diagnosis (31).…”

“…Furthermore, this resistance is associated with the activation of Stat3 and EMT [19,57]. The molecular interaction of STAT3 and Snail, ZIP6, and also the other transcription factors suggests that it is part of a core that controls EMT in normal and pathological conditions [58,59]. These data highlight the central role of STAT3, which could suggest that it could be a therapeutic target, its inhibition leading to a decrease in the main characteristics of tumor cells: proliferation, motility, invasion and upregulation of apoptosis.…”

“…Consequently, the five-year survival rate is low, ranging from 5 to 15% (Yu et al, 2009;Hsu et al, 2012). In 2008, the incidence rate of gastric cancer (0.738 million mortalities and 0.98 million new cases) ranked it as the second leading cause of cancer all over the world (Jemal et al, 2011).…”
